| Objective: Based on biomedical big data,key genes of HP-related chronic atrophic gastritis were screened to explore its pathogenesis and the influence of An Wei Decoction on it.Methods: The chip data related to HP-associated chronic atrophic gastritis was downloaded from Gene Expression omnibus(GEO)database,and the differential genes of HP-associated chronic atrophic gastritis were screened by GEO2 R software.Bioinformatics analysis was conducted,and the core proteins were determined according to the number of protein participating pathways.PCR and Western blot were used to detect the effect of An Wei decoction on its expression.Results: After retrieval and analysis of GSE13873 and GSE27411 series chip data,20 differential genes at the intersection of the two chips were taken for bioinformatics analysis,and it was found that APOA1,CD36 and CFTR might be the core proteins of the signaling pathway.Western blot and PCR results showed that the expression of CD36 protein was down-regulated(P <0.05),and the expression of APOA1 and CFTR protein were up-regulated(P< 0.05)in the gastric tissue of rats with HP-associated chronic atrophic gastritis.Compared with the model group,the expression of CD36 protein in the high-dose group,medium-dose group and low-dose An Wei decoction group was up-regulated,and the expression of APOA1 and CFTR protein was down-regulated(P < 0.05),which was closely related to the incidence of HP-related chronic atrophic gastritis.Conclusions: Chronic atrophic gastritis in GEO Hp correlation chip bioinformatics analysis combined with Western blot and PCR further validation showed APOA1,CD36,CFTR protein,which is the core of the signal path experimental results are consistent with the bioinformatics,at the same time give Anwei Decoction to interfere,find Anwei Decoction intervention can significantly improve the CAG rats stomach tissues CD36,APOA1 and the expression of CFTR,and gastric acid secretion,digestion and absorption of fat,PPAR signaling pathways,AMPK pathway. |
