Objective:Based on NLRP3 inflammatory body pathway,Weifu kang granule was used to interfere with Helicobacter pylori associated gastritis(Helicobacter pylori associated gastritis,Hp AG)mice,and to explore the mechanism of Weifukang granule on Hp AG mice.Methods:Fifty-two SPF BALB/c mice were randomly divided into normal group(10 mice)and modeling group(42 mice).The normal group was given distilled water and the model group was given Hp suspension.Control group and modeling group were given gavage once every other day for 5 times.4 weeks after the last intragastric administration,2 mice were randomly selected from the modeling group and sacrificed to take the gastric mucosa tissue for disease examination for model identification(part of the mice were stained by HE and part were tested by rapid urease test,and if both were positive,the modeling was successful).After successful modeling,the remaining 36 mice in the modeling group were randomly divided into model group,traditional Chinese medicine group,Western medicine group and integrated Chinese and Western medicine group.The control group and model group were given normal saline0.4ml/ piece intragastrically,the traditional Chinese medicine group was given Weifukang granules 0.4ml/ piece intragastrically,the western medicine group was given Western medicine quartet 0.4ml/ piece intragastrically,and the integrated Chinese and Western medicine group was given Weifukang granules and Western medicine quartet 0.4ml/piece intragastrically,once a day for 2 weeks in each group.During the experiment,the mental state,body weight,hair color,food intake and urine of the mice were observed.After the last administration of the drug,all mice were sacrificed after fasting for 12 h.The gastric mucosa histopathology of the mice was observed by HE staining,and the expression levels of IL-1β and IL-18 in the gastric mucosa of Hp AG mice were detected by Elisa.m RNA expression levels of NLRP3,ASC and caspase-1 in gastric mucosa of mice were detected by q RT-PCR.Results:(1)General conditions of mice: compared with the normal group,the mental state of mice in the model group was poor,the hair was rough and dark,the hair was prickly and dull or poor,the activity level was significantly reduced,lethargy,yellow urine,loose stool,and the weight growth slowed down.Compared with the model group,the mental state,body weight,hair color,water intake,urine and feces of mice in the Chinese medicine group,Western medicine group and integrated Chinese and Western medicine group were improved after intragastriction,and the integrated Chinese and western medicine group had the best improvement.(2)HE staining: Compared with the normal group,the gastric mucosa in the model group was significantly congested and swollen,the mucosa was thinner and even sunken,the cell morphology and structure were abnormal,the number of epithelial cells was reduced,the gastric pits were clear,the glandular ducts were neatly arranged,the number of glandular cells was reduced,and a large number of inflammatory cells such as plasma cells,lymphocytes and neutrophils were observed.After drug intervention,Compared with the model group,the gastric mucosa inflammation of mice was improved,and the improvement effect of integrated Chinese and western medicine group was the most obvious.(3)Elisa results: Compared with normal group,the expressions of IL-1β and IL-18 in model group were significantly increased(P<0.01).Compared with model group,the expressions of IL-1β and IL-18 in TCM group,Western medicine group and integrated Chinese and Western medicine group were significantly decreased(P<0.01).Compared with the western medicine group,the IL-1β of the Chinese medicine group was increased(P < 0.05),but the IL-18 of the Chinese medicine group was not significantly different(P > 0.05),and the IL-1β and IL-18 of the integrated Chinese and western medicine group were significantly decreased(P < 0.01).Compared with the traditional Chinese medicine group,the expressions of IL-1β and IL-18 in the integrated Chinese and western medicine group were significantly decreased(P<0.01).(4)qRT-PCR results: Compared with normal group,m RNA expressions of NLRP3,ASC and caspase-1 in gastric mucosa tissues of mice in model group were significantly increased(P < 0.01).Compared with model group,m RNA expressions of NLRP3,ASC and caspase-1 in TCM group,Western medicine group and integrated Chinese and Western medicine group were decreased(P < 0.01 or P < 0.05).Compared with western medicine group,m RNA expressions of ASC and caspase-1 in gastric mucosa tissue of mice in Chinese medicine group had no statistical difference(P > 0.05),NLRP3,ASC and caspase-1 in integrated Chinese and western medicine group and NLRP3 and Caspase-1 in Chinese medicine group were decreased(P < 0.01 or P <0.05).Compared with the traditional Chinese medicine group,the m RNA expressions of NLRP3,ASC and caspase-1 in the integrated Chinese and western medicine group were significantly decreased(P < 0.01).Conclusion:Weifukang granules can improve the gastric mucosal inflammatory response of Hp AG mice,and has a good therapeutic effect on Hp AG mice.The mechanism of its therapeutic effect on Hp AG mice may be to down-regulate the expression levels of ASC,caspase-1 and inflammatory factors IL-1 β and IL-18 by regulating the NLRP3inflammator-body pathway,so as to reduce the inflammatory response.To achieve the purpose of treating Hp AG,and combined with PPI Western medicine quadruple therapeutic effect is better than simple Chinese medicine group and western medicine group. |
