| Objective: To observe the effect of Zhu Lian stimulant type I acupuncture on the expression of brain-derived neurotrophic factor(BDNF),its receptor Trk B and tissue homogenate cyclic adenosine phosphate(cAMP)in rats with diabetic bladder,and to explore the mechanism of Zhu Lian stimulant type I acupuncture on DNB.Methods: Fifty SD rats were randomly divided into control group,model group,western medicine group,ordinary acupuncture group and Zhu Lian stimulant type Ⅰ acupuncture group(type Ⅰ treatment group),with 10 rats in each group.DNB rat model was prepared by intraperitoneal injection of streptozotocin(STZ).The western medicine group was given mecobalamine tablet intragastric administration;In the ordinary acupuncture group,"Zhongji","Sanyinjiao","Lieqi" and "Taichong" points were selected,and the manipulation of leveling and replenishing and purging was performed.The type Ⅰ treatment group(the same as the ordinary acupuncture group)was treated with Zhulian stimulation type Ⅰ acupuncture;Sampling materials were collected after 4 weeks of treatment.The wet weight of bladder in each group was compared.The morphology of rat bladder was observed by hematoxylin and eosin staining.The expression level of BDNF protein in rat bladder was detected by immunohistochemistry.Western Blot(WB)was used to detect cAMP protein expression in rat bladder.The expression level of TrKB protein in rat bladder was determined by ELISA.Real-time fluorescence quantitative PCR(RT-PCR)was used to detect the expression levels of BDNF mRNA and cAMP mRNA in rat bladder group.Results: Compared with control group,wet weight of bladder in model group was increased(P<0.01).BDNF and mRNA,cAMP and mRNA and TrKB were decreased(P<0.01).Compared with model group,the wet weight of bladder in western medicine group,common acupuncture group and Zhu Lian acupuncture Ⅰ group decreased(P<0.01);The expression levels of BDNF,mRNA and cAMP mRNA were increased(P<0.01),and the expression levels of TrKB and cAMP were increased(P<0.05).Compared with the ordinary acupuncture group,the wet weight of bladder in the western medicine group and Zhu Lian acupuncture Ⅰ group was decreased(P<0.05),the expression levels of BDNF mRNA,cAMP mRNA and TrKB were increased(P<0.01),and the expression levels of BDNF and cAMP were increased(P<0.05).There was no significant difference in bladder wet weight,BDNF,mRNA,Trk B,cAMP and mRNA between western medicine group and Zhu Lian acupuncture group.Compared with the model group,the bladder structure of rats in the western medicine group and the common acupuncture group was improved,and the Zhu Lian stimulation-type Ⅰ acupuncture group had the best improvement.Conclusion:1.Zhu Lian excitation-type I acupuncture,mecobalamine and ordinary acupuncture can reduce the wet weight of bladder,Zhu Lian excitation-type I acupuncture and mecobalamine are superior to ordinary acupuncture;2.Zhu Lian stimulant type I acupuncture,mecobalamine and ordinary acupuncture can effectively improve the bladder structure,Zhu Lian stimulant type I acupuncture group is better than mecobalamine and ordinary acupuncture;3.The mechanism of Zhu Lian stimulant type I acupuncture in the treatment of diabetic neurogenic bladder may be related to the up-regulation of BDNF protein and mRNA,Trk B protein,cAMP protein and mRNA expression. |
