| Objective: To exposit the efficacy of Ning Xue Hua Ban Decoction on primary immune thrombocytopenia(ITP)rats,and probe into the study of immunomodulatory mechanism of Ning Xue Hua Ban Decoction on ITP rats by regulating the expression of Treg through JAK3/STAT5 pathway.Methods: Rats were randomly divided into blank group,model group,Ning Xue Hua Ban Decoction’s low-dose group,Ning Xue Hua Ban Decoction’s medium-dose group,Ning Xue Hua Ban Decoction’s high-dose group,prednisone group.ITP rat model was established by intraperitoneal injection of rabbit antirat platelet antibody serum(APS),except for the blank group.Drug administration began on the 7th day of modeling.Except for blank group and model group,the rats were given normal saline intragastric administration,and the other groups were given corresponding drugs for 14 consecutive days.All rats were killed after drug administration on the 14 th day.The changes of platelets and white blood cells in each group were evaluated before the modeling,on the 7th day after modeling,and the end-of-treatment.After the end of treatment,the number of megakaryocytes was counted on the bone marrow smear of one side of the femur,and the m RNA expression of JAK3,STAT5 b and Foxp3 in peripheral blood was detected by RT-PCR.Results:(1)On two days after APS injection,bleeding spots were scattered throughout the body on modeling rats.After treatment,the bleeding point was obviously subsided.(2)On the 7th day after modeling,the number of platelets in all modeling groups was significantly lower than that in the blank group,with statistical significance(P<0.05).(3)After treatment:(1)Platelet: the number of platelets in all of drug treated groups increased significantly compared with the model group(P<0.05),but there were no significant difference between the Ning Xue Hua Ban Decoction groups and the prednisone group(P>0.05),and the increase was greater in the Ning Xue Hua Ban Decoction medium-dose group.(2)megakaryocyte: the number of megakaryocytes in the model group was significantly less than that in the blank group(P<0.05).Compared with the model group,the number of megakaryocytes in bone marrow in all of drug treated groups were increased(P<0.05),and the number of megakaryocytes in the Ning Xue Hua Ban Decoction medium-dose group was highest in all of groups.(3)JAK3,STAT5 b,Foxp3 m RNA expression: all model groups were significantly lower than the blank group(P<0.05),the all of drug treated groups’ s expression were increased,and the JAK3 and STAT5 b expression quantity in prednisone group is lower than the Ning Xue Hua Ban Decoction’s low-dose group,Ning Xue Hua Ban Decoction’s medium-dose group,Ning Xue Hua Ban Decoction’s high-dose group(P<0.05),the JAK3,STAT5 b,Foxp3 expression in Ning Xue Hua Ban Decoction’s low-dose group was lower than in Ning Xue Hua Ban Decoction’s medium-dose group and Ning Xue Hua Ban Decoction’s high-dose group(P<0.05),the JAK3 and STAT5 b expression in Ning Xue Hua Ban Decoction’s medium-dose group is lower than the Ning Xue Hua Ban Decoction’s high-dose group(P>0.05),and the Foxp3 expression was higher than Ning Xue Hua Ban Decoction’s high-dose group(P>0.05).Conclusions:(1)Treatment of Ning Xue Hua Ban Decoction on ITP rat can increase the number of platelets in blood and the number of megakaryocytes in bone marrow,and can effectively improve the bleeding symptoms.(2)Ning Xue Hua Ban Decoction can upregulate the expression of JAK3,STAT5 b,Foxp3 m RNA and promote the immunosuppressive function of CD4+CD25+Treg cells in ITP rats.(3)Ning Xue Hua Ban Decoction’s medium-dose group and Ning Xue Hua Ban Decoction’s high-dose group were superior to the Ning Xue Hua Ban Decoction’s low-dose group. |
