Objective: Based on data mining technology,to analyze the prescription medication of modern doctors in the treatment of premature ovarian failure with traditional Chinese medicine,in order to explore the medication rule of traditional Chinese medicine in the treatment of POF and summarize the core drugs.The potential mechanism of action of core drugs was explored by using the network pharmacology method in order to provide theoretical basis for the treatment of POF with traditional Chinese medicine.Methods: 1.Data mining research: collect the documents of traditional Chinese medicine treatment of POF in line with the standards of Naturalization and Exclusion from January 1,2000 to November 30,2022,collect the CNKI database,Wanfang Data database,VIP database,and CBM database,and use Excel to establish the database.After standardized processing,enter it into the auxiliary platform of traditional Chinese medicine inheritance(V2.50),Use data mining technology to analyze the characteristics of TCM syndrome types and prescription medication,explore common drug combinations,refine core prescriptions,and summarize the medication rules of TCM in the treatment of POF.2.Network pharmacology research: select the core prescription obtained from data mining to conduct network pharmacology research.First of all,use the database of TCMSP to obtain the effective drug components of the core prescription,use Pubchem,Swiss Target Prediction database and Uni Prot database to obtain the target genes of the effective drug components,and use the Cytascape 3.7.2 software to construct the network diagram of "core drug-effective component-target gene";Then use Gene Cards,Dis Ge Net and Drugbank databases to obtain POF-related disease genes;Then,the core drug target gene and disease gene are respectively introduced into the jvenn platform to obtain the intersection gene of the two,and the Wayne map is made;Then,the intersection gene is imported into the String database to build the protein-protein PPI network relationship;Finally,the intersection gene was imported into Metascape database and David database for GO and KEGG enrichment analysis to explore the mechanism of action of core drugs.Results: 1.Data mining research: A total of 114 eligible documents were screened,including 114 prescriptions,108 traditional Chinese medicines,and12 TCM syndrome types.According to the frequency,the TCM syndrome types are kidney deficiency,kidney deficiency and blood stasis,kidney deficiency and liver depression,liver and kidney yin deficiency,kidney deficiency and blood deficiency,spleen and kidney yang deficiency,heart and kidney failure,etc;The top 10 traditional Chinese medicines with the highest frequency of drug use are Angelica sinensis,Cuscuta chinensis,Radix Rehmanniae Preparata,Fructus Lycii,Epimedium,Chinese yam,Paeonia lactiflora,Cornus officinalis,Glycyrrhiza uralensis and Salvia miltiorrhiza;The top ten drugs with the highest frequency of efficacy classification are respectively tonifying deficiency,activating blood circulation and removing blood stasis,clearing heat,astringent,calming nerves,regulating qi,relieving external symptoms,warming the interior,promoting water and dampness,calming the liver and calming the wind;From high to low,the proportion of the four gases of drugs is warm,calm,cold,cool and hot;The proportion of the five flavors of drugs from high to low is sweet,spicy,bitter,sour,salty and astringent;The proportion of drug meridians from high to low is liver meridian,kidney meridian,spleen meridian,heart meridian,lung meridian,stomach meridian,gallbladder meridian,large intestine meridian,pericardium meridian,bladder meridian,triple energizer meridian and small intestine meridian.A total of 35 commonly used drug combinations were obtained based on association rule statistics,and the first five groups with the highest frequency were Cuscuta chinensis and Angelica sinensis group,Radix Rehmanniae Preparata and Cuscuta chinensis group,Radix Rehmanniae Preparata and Angelica sinensis group,Cuscuta chinensis and Lycium barbarum group,Angelica sinensis and Lycium barbarum group;A total of 61 groups of closely related associations were obtained,of which 30 groups were the combination of two drugs,and the highest confidence was Cornus officinalis and Rehmannia officinalis;There are 25 groups of combinations of three drugs,with the highest confidence level of white peony,wolfberry fruit and angelica;There are 6 groups of four drug combinations,with the highest confidence level being Radix Rehmanniae Preparata,Angelica Sinensis,Fructus Lycii and Fructus Cuscuta,which are the core prescriptions.Based on entropy cluster analysis,8 groups of new prescriptions were obtained,namely:(1)Schisandra chinensis,Chuanxiong,Leonurus heterophyllus and Astragalus membranaceus;(2)Raspberry,dodder,wolfberry,peach kernel,raw rehmannia glutinosa;(3)Ligusticum chuanxiong,cornus officinalis,Chinese yam,mulberry,dendrobium;(4)Safflower,Radix Pseudostellariae,Herba Eclipta,Radix Paeoniae rubra;(5)Cornus officinalis,rehmannia officinalis,peony bark,eucommia ulmoides;(6)Sour jujube kernel,floating wheat,angelica,glutinous rice root;(7)Tortoiseshell,antler glue,atractylodes macrocephala,rose;(8)Ligustrum lucidum,Morinda officinalis,Eclipta chinensis,antler gum,orange peel.2.Network pharmacology research: A total of 51 effective components,339 potential action targets and 1653 disease targets of POF were selected from the core drugs for the treatment of POF.A total of 114 intersection genes were obtained after Wayne mapping.Network analysis shows that the core drugs include quercetin β-Fifty-one active ingredients including beta-sitosterol and stigmasterol,the core targets are AKT1(protein kinase B),TNF(tumor necrosis factor),ESR1(estrogen receptor 1),EGFR(epidermal growth factor receptor),etc.,mainly through PI3K-Akt signaling pathway,VEGF signaling pathway Estrogen signaling pathway,HIF-1signaling pathway,ovarian steroidogenesis,etc.play a role.Conclusion: The results of data mining show that the treatment of POF with traditional Chinese medicine is mainly kidney tonifying.Angelica sinensis,dodder seed,rehmannia glutinosa and wolfberry fruit can be used as the core drugs for POF treatment,providing some ideas for the standardization and internationalization of the treatment of POF with traditional Chinese medicine;The results of network pharmacology show that the core drug treatment of POF has the characteristics of multi-component coaction,multi-target and multi-path co-regulation,which provides a certain theoretical basis for the scientific research and clinical application of POF.
