Objective:Data mining was used to preliminarily explore Professor Fubin’s clinical experience in treating active RA and to induce his pastime medication pattern.High frequency drugs and core prescriptions derived from data mining were analyzed by network pharmacology to speculate on their mechanism of action in the treatment of active RA,which may provide the basis for the next clinical validation of TCM’s efficacy.Methods:1.To collect the active phase RA cases treated in outpatient clinic and ward of Professor Fubin,standardize the TCM in prescriptions,and establish a database.Microsoft Excel,SPSS software was used as the data mining tool to count the frequency,four Qi,five flavor and GUI Jing of TCMs and mine their common drugs,drug pairs and core prescriptions to explore the pastime medication pattern of Prof.Fubin in treating RA.2.The active chemical constituents of Radix Achyranthes Bidentatae,Rhizoma Notopterygii,Radix Paeoniae Alba,Radix Atractylodis macrophyllae,semen coix seed,Radix Scutellariae,Radix Bupleuri,cortex angelicae sinensis,Radix Glycyrrhizae,Radix Scutellariae,Lonicera japonica,and Radix deer were collected and retrieved by the traditional Chinese medicine systems pharmacology analysis platform(tcmsp),and the active compounds and corresponding targets were retrieved by batman-tcm analysis tool.Currently known disease targets associated with rheumatoid arthritis were retrieved through genecards and OMIM databases with the keywords " rheumatoid arthritis,RA ".The intersection target genes of core prescription and RA disease target were calculated by R language,and the intersection genes of disease and drug were entered into string database to construct the PPI network and the core targets were screened out.Go functional enrichment analysis of intersection target genes was performed with the Encyclopedia of genomes(KEGG)metabolic pathway enrichment analysis by R language to illustrate the role of target proteins in gene function.Results:1.The commonly used traditional Chinese medicines(TCM)are Angelica sinensis,Glycyrrhiza uralensis,Lonicera cartilagina,Rhizoma Notopterygii,Radix Paeoniae Alba,Radix Atractylodis Macrocephalae,cortex coix seed,and cortex gallinaceae.The efficacy of traditional Chinese medicines(TCM)mainly involves clearing heat,removing wind and dampness,and toning deficiency.From the four gas statistical results,cold cooling drugs were used the most,warm drugs and peace drugs the second time.From the five flavor statistics,bitter traditional Chinese medicine was used the most,followed by Gan,Xin,salty,astringent and acidic,and an association analysis concluded that the medicinal pair included Qiang → alone,Huangbai →Baizhu,Danggui → deer Tiao,forsythia → Jinyinhua.The core prescription is:Jinyinhua,caprifolia vine,whole scorpion,Aconitum carmichaeli snake,deer Tiao root,Angelica sinensis,Scutellaria baicalensis root,Bupleurum chinense root,Chenpi root,and Gancao radix.2.The main components through which core prescriptions come into play areβSitosterol,stigmasterol,kaempferol,luteolin,quercetin,methyl palmitate,etc;The core targets of corepressors were: TP53,AKT1,CASP3,TNF,and CCND1;Core prescribing is regulated by biological processes such as cellular response to chemical stress,response to lipids,response to reactive oxygen species,regulation of DNA binding transcription factor activity,response to bacteria,through protein kinase activity,DNA binding transcription factor binding,RNA polymerase II specific DNA binding,transcription factor binding,protein serine / threonine kinase activity Chromatin binding and so on,targeting PI3 K Akt,MAPK,FOXO,HIF-1,AGE-RAGE and other signaling pathways to exert therapeutic effects against RA.Summary:1.Professor Fu Bin The commonly used drugs for the treatment of RA in the active stage are Angiocarpus persicum,Phellodendron chinensis,Atractylodes atractylodes,Angelica sinensis,Forsythia forsythia,Honeysuckle2.Professor Fu bin’s core prescriptions for treating active RA are: honeysuckle,honeysuckle vine,scorpion,blacktail snake,herbivore,angelica,scutellaria,bupleurum,tangerine peel,licorice.3.Core prescription may affect PI3 K Akt signaling pathway,regulate inflammatory response,block inflammatory "positive feedback" of disease,prevent tumor like degeneration of synovial cells,and play an anti rheumatic role.It is speculated that core prescription may inhibit cell proliferation and inflammatory response by affecting gene expression,protein synthesis and enzyme activity,so as to treat active RA. |
