| Objective: 1.To explore the expression of Lnc RNA H19 in peripheral blood of patients with spontaneous intracerebral hemorrhage(SICH),and to evaluate the diagnostic value of Lnc RNA H19 in SICH.2.To explore the correlation between the expression level of Lnc RNA H19 and the severity and inflammatory factors,severity and prognosis in patients with SICH.3.To analyze the effect of Lnc RNA H19 in patients with SICH by regulating apoptosis-related factors.Methods: The research method is divided into two parts.Clinical research part: 1.60 patients with SICH diagnosed(case group)in Tai zhou People’s Hospital from December 2021 to June 2022 and 30 physical examination persons(control group)were included.Collect general data:age,gender,smoking history,drinking history,hypertension history,hyperlipidemia history,diabetes history,collect laboratory data:bleeding and coagulation,white blood cell count,neutrophil count,etc.2.The expression of Lnc RNA H19 in peripheral blood of was detected by RT-q PCR.ROC curve was drawn to evaluate the diagnostic value of Lnc RNA H19 for SICH.3.The levels of interleukin-6(IL-6)and procalcitonin(PCT)were detected by enzyme-linked immunosorbent assay(ELISA),and the correlation between the expression level of Lnc RNA H19 and WBC,NEUT%,IL-6,PCT in patients with SICH was analyzed by Pearson correlation.4.The Glasgow Coma Scale(GCS)score at admission and Glasgow Outcome Scale(GOS)score at 3 months after SICH were recorded.Pearson correlation was used to analyze the correlation between the expression level of Lnc RNA H19 in patients with SICH and GCS score and GOS score.Cell experiment part:1.Human umbilical vein endothelial cells were cultured in vitro and transfected into groups:Lnc RNA H19-si RNA transfection group,transfection mimic negative control group(si RNA NC group)and blank control group(Control group)was constructed..The expression of Lnc RNA H19 in each group was compared.2.Flow cytometry was used to detect apoptosis in each group.3.The expression levels of apoptosis-related factors Bcl-2,Bax and Caspase-3 in each group were detected by Western blot.Results:1.Clinical research results:1)The expression level of Lnc RNA H19 in peripheral blood of patients with SICH was higher than that in healthy controls(3.07 ± 1.39 vs.1.26 ± 1.01,P < 0.05).The ROC curve showed that the AUC of Lnc RNA H19 was 0.882,the sensitivity and specificity were 0.850 and 0.900,respectively,and the optimal cutoff value was 1.830.2)The level of Lnc RNA H19 in peripheral blood of patients with SICH was positively correlated with WBC(r=0.313,P<0.05)and IL-6(r=0.400,P<0.05).There was no significant correlation with NEUT%(r=0.064,P>0.05)、PCT(r=0.192,P>0.05)3)The level of Lnc RNA H19 in peripheral blood of patients with SICH was negatively correlated with GCS score(r=-0.436,P<0.05).There was no significant correlation with GOS score(r=0.243,P>0.05).2.Cell experiment results :1)The expression level of Lnc RNA H19 in Lnc RNA H19-si RNA transfection group was significantly lower than that in Control group and si RNA NC group(0.38±0.16 vs.1.00±0.12 vs.1.01±0.12,P < 0.05).2)The apoptosis rate of Lnc RNA H19-si RNA transfection group was significantly lower than that of Control group and si RNA NC group(7.10%±0.35% vs.12.68%±0.24% vs.12.07%±0.63%,P<0.05).2)At the m RNA level,The gene expression levels of Bax in Lnc RNA H19-si RNA transfection group were significantly lower than those in Control group and si RNA NC group(0.35±0.06 vs.1.01±0.08 vs.1.03±0.08,P<0.05).The gene expression levels of Caspase-3 in Lnc RNA H19-si RNA transfection group were significantly lower than that in the other two groups(0.36±0.04 vs.1.01±0.09 vs.0.99±0.04,P<0.05).The gene expression levels of Bcl-2 in Lnc RNA H19-si RNA transfection group were significantly higher than that in the other two groups(3.85±0.30 vs.0.99 ± 0.09 vs.0.97±0.16,P<0.05).3)At the protein level,the protein expression levels of Bax in Lnc RNA H19-si RNA transfection group were significantly lower than those in Control group and si RNA NC group(0.26±0.06 vs.1.01±0.06 vs.1.02±0.03,P<0.05).The protein expression levels of Caspase-3 in Lnc RNA H19-si RNA transfection group were significantly lower than that in the other two groups(0.26±0.06 vs.1.01±0.06 vs.1.02±0.03,P<0.05).The protein expression levels of Bcl-2 in Lnc RNA H19-si RNA transfection group were significantly higher than that in the other two groups(3.65±0.27 vs.1.01 ± 0.14 vs.0.95±0.17,P<0.05).Conclusions:1.The expression level of Lnc RNA H19 in peripheral blood of patients with SICH was significantly higher than that in the healthy group,suggesting that Lnc RNA H19 can be used as an auxiliary diagnostic marker for SICH.2..The expression level of Lnc RNA H19 in peripheral blood of patients with SICH was positively correlated with the expression of WBC and IL-6,suggesting that Lnc RNA H19 may participate in the pathological process of SICH by regulating inflammatory response.3.Lnc RNA H19 in peripheral blood of patients with SICH is related to the severity of the disease.4.The results of in vitro cell experiments show that Lnc RNA H19 can participate in the pathological process of SICH by regulating apoptosis,and which could further provide experimental basis for the related mechanism of the pathological process of SICH. |
