Objective: based on the method of data mining and analysis,to explore the prescription rule of traditional Chinese medicine in the treatment of chronic atrophic gastritis(CAG)of liver stomach qi stagnation type in the Department of spleen stomach hepatobiliary Diseases of Jinan traditional Chinese Medicine Hospital.This thesis summarizes the medication ideas,rules,characteristics and core Chinese medicine in the treatment of CAG,and uses the method of network pharmacology to find the mechanism of Action of core Chinese medicine in the treatment of CAG.Methods: 1.The electronic medical records of inpatients with CAG of liver and stomach qi stagnation type were collected from December 2020 to January 2023 in the Department of spleen,stomach,Hepatobiliary Diseases,Jinan Traditional Chinese Medicine Hospital.Input the data of patients who meet the inclusion criteria into Excel database,and then use SPSS data analysis software to analyze the prescriptions of Chinese medicine by statistical analysis,Association rule analysis and cluster analysis were used to obtain high frequency Chinese medicine and its taste,meridian tropism,function and combination law.2.The core Chinese medicine for the treatment of CAG of liver and stomach qi stagnation type was selected for network pharmacology research,and the active components and targets of the core Chinese medicine were screened by traditional Chinese medicine system pharmacological database and analysis platform(TCMSP).Then the disease-related targets were screened by Gene Cards database and OMIM database.Obtain the intersection targets of drugs and diseases in the Venny2.1.0 online software and draw the Wayne diagram.The drug-target-disease network map was constructed based on the relationship between the active ingredient of the core Chinese medicine and the target of CAG by Cytoscape3.8.2 software.The intersection targets of drugs and diseases are inputted into the STRING11.0 online database to obtain the data of protein-protein interactions.The data of protein-protein interactions are analyzed by Cytoscape3.8.2software to get the core targets.The last intersection targets import drugs and disease Metascape database for GO enrichment and KEGG pathway enrichment analysis.Results: 1.This study included the prescriptions of 200 inpatients for the first visit,involving 158 kinds of Chinese medicine.among them,Poria cocos,Amomum villosum,Pinellia ternata,Radix Paeoniae Alba,Bupleurum,Atractylodes macrocephala,Chicken Nei Jin,Shenqu,Yujin,Xiangfu and Muxiang are among the top 10.Among the four qi of drugs,the frequency of warm drugs was the highest,followed by flat and cold.Among the five flavors,pungent medicine is the most frequently used,followed by sweet taste and bitter taste.Drug meridians mainly belong to spleen and stomach meridians,followed by lung and liver meridians.The drug combinations with higher association rules are Pinellia ternata,Amomum villosum-Poria cocos,Atractylodes macrocephala,Amomum villosum-Poria cocos,Amomum villosum-Amomum villosum,Radix Paeoniae Alba-Radix Paeoniae Alba.Six groups of drug combinations were obtained by cluster analysis: cuttlebone,Evodia,Rhizoma Coptidis;Rhizoma Corydalis,Radix Paeoniae Alba,Radix Bupleuri,turmeric,wood incense,Amomum villosum;Fructus Aurantii,betel nut;cardamom,patchouli,tangerine peel,Coix seed;Poria,Atractylodes macrocephala;yam,lentil,Euryale,Pueraria lobata,Radix Astragali.Comprehensive analysis showed that Poria cocos,Amomum villosum,Pinellia ternata,Radix Paeoniae Alba,Bupleurum and Atractylodes macrocephala were the core Chinese medicine 2.In this study,through the analysis of the mechanism of core Chinese medicine in the treatment of CAG,50 active components of core Chinese medicine were found.the main active components are quercetin,kaempferol,β-sitosterol,baicalein,stigmasterol,isorhamnetin and so on.The core targets for the treatment of chronic atrophy are AKT1,IL6,TNF-α,IL1 B,HIF-1 α,PTGS2,MMP9,NF-κ B and so on.GO enrichment analysis showed that it was mainly involved in biological processes such as response to inorganic substances,response to heterogeneous stimuli,response to lipopolysaccharide,positive regulation of cell death,regulation of apoptosis signal pathway and molecular functions such as protein homodimerization activity,DNA binding transcription factor binding,kinase binding,cytokine receptor binding and so on.The cell composition mainly involves the membrane raft,the vesicle cavity,the side of the membrane,the outer membrane of organelles and so on.KEGG enrichment analysis mainly revolve NFkappa B signal pathway and HIF-1 signal pathway.Conclusion: 1.CAG of liver and stomach qi stagnation type is mainly revolved to spleen,stomach and liver.the treatment of CAG is mainly related to spleen,spleen and liver.The treatment principles is gasification and dampness,invigorating the spleen and stomach,and taking warm and smooth as the basic tone when forming prescriptions.Most of the drugs with spicy and bitter taste are selected,and the drugs for promoting qi and invigorating the spleen are mainly used.At the same time,the dialectical compatibility of tonic drugs,digestible drugs,and bloodactivating drugs is dialectica Poria cocos,Amomum villosum,Pinellia ternata,Radix Paeoniae Alba,Radix Bupleuri and Atractylodes macrocephala.2.Quercetin,kaempferol,β-sitosterol,baicalein,stigmasterol,isorhamnetin and other active components of core Chinese medicine play a therapeutic role in inhibiting inflammation,protecting gastric mucosa,regulating mood and preventing carcinogenesis through core targets such as AKT1,IL6,TNF-α,IL1 B,HIF-1α,PTGS2,MMP9,NF-κB and regulating NF-kappa B signal pathway and HIF-1 signal pathway.This study provides a train of thought and theoretical basis for the clinical treatment and mechanism of chronic atrophy of liver and stomach qi stagnation.
