| Objective The expression level of Cysteinyl leukotriene 1 receptor(Cys LTR1)and Leukotriene C4(Leukotriene)were detected in neutrophils,eosinophils and basophils C4,LTC4)was detected in Allergic Asthma(AA),Allergic Rhinitis(AR),AR combined with AA,ARA,and Non-Allergic asthma The expression level in blood of patients with Asthma(NAA)is discussed to discuss its role and significance in the course of disease pathogenesis,and the level of Fe NO is compared to further explore the causes of inter-disease heterogeneity.Methods A total of 78 volunteers(17 Healthy Control subjects(HC),21 AA patients,28 AR patients,6 ARA patients and 6 NAA patients)were recruited.General data and venous blood from the elbow were collected.Flow cytometry was used to obtain Cys LTR1 expression levels on neutrophils,eosinophils and basophils of the volunteers.Enzyme-linked immunosorbent assay(ELISA)was used to measure plasma levels of LTC4 inflammatory factors in volunteers and to assess exhaled nitric oxide(Fractional exhaled sclerosis)(3)concentration of oxide(Fe NO),and the correlation between plasma LTC4 and Fe NO and the expression of three granulocyte receptors was analyzed.Results 1.Flow cytometry results: Compared with HC group,the expression of Cys LTR1 on neutrophils,eosinophils and basophils of allergic patients in AA,AR and ARA groups was increased,and the difference was statistically significant.The expression of Cys LTR1 on neutrophils and basophils in NAA group increased,and the difference was statistically significant,while the expression of Cys LTR1 on eosinophils in NAA group had no significant change,and the difference was not statistically significant.Compared with NAA group,the expression of Cys LTR1 on eosinophils in AA,AR and ARA allergic patients was increased,and the difference was statistically significant.2.ELISA results: The levels of LTC4 in AA,AR and ARA allergic patients were significantly higher than those in HC and NAA groups,with statistical significance(P < 0.05),while the levels of LTC4 in NAA group were not significantly different from those in HC group(P > 0.05).3.Fe NO results: Fe NO level in AA,AR,ARA and NAA groups was higher than that in HC group;Fe NO level in AA and ARA groups was significantly higher than that in NAA group;Fe NO level in ARA group was higher than that in AA,NAA and AR groups,and the difference was statistically significant(P < 0.05).4.Correlation between plasma LTC4,Fe NO and expression of three kinds of receptors on granulocytes: In allergic patients with AA,AR and ARA,Cys LTR1 expressed on eosinophils was positively correlated with Fe NO,while Cys LTR1 expressed on eosinophils in NAA patients was not correlated with Fe NO.There was no correlation between the expression of Cys LTR1 on eosinophils and plasma LTC4 in AA,AR,NAA and ARA groups.There was no significant correlation between the expression of Cys LTR1 on neutrophils and basophils and LTC4 and Fe NO in the AA,AR,NAA and ARA groups.Conclusion1.Neutrophils,eosinophils and basophils may be involved in the pathogenesis of AA,AR and ARA allergic diseases through Cys LTR1.Neutrophils and basophils may be involved in the pathogenesis of NAA through Cys LTR1.2.LTC4 is involved in the pathogenesis of AA,AR and ARA3.LTC4 and Cys LTR1/ eosinophilic granulocyte may be markers of inflammatory differences between NAA and AA,AR,ARA and other allergic diseases,and may be the cause of the differentiation between NAA and AA,AR,and ARA.4.Fe NO is an auxiliary diagnostic method for asthma and allergic rhinitis,which is of great significance in distinguishing allergic asthma from non-allergic asthma.Fe NO is positively correlated with Cys LTR1 expressed on eosinophils of allergic diseases such as AA,AR and ARA. |
