Objective:To observe the clinical efficacy of Wenxin granule in the treatment of chronic coronary syndrome with ventricular premature contraction of qi and yin deficiency syndrome on the basis of conventional western medicine treatment,and to study the network pharmacological mechanism of Wenxin granule monarch drug party participating in the minister drug Huang Jing.Methods:1.Clinical research : 59 medical records that met the inclusion criteria were randomly assigned to the treatment group and the control group by random sampling.The control group was given conventional western medicine anti ventricular premature beat drugs.The treatment group was treated with Wenxin granule on the basis of the control group.The efficacy was evaluated before and after treatment.2.Network pharmacology research : TCMSP,Gene Cards,OMIM and other databases and Metascape platform were used to screen the active components and potential targets of Dangshen-Huangjing in the treatment of premature ventricular contraction.Cytoscape 3.9.1 software and STRING platform were used to construct ’ drug-component-target network diagram ’ and ’ PPI network diagram ’,and core targets were screened out.GO function and KEGG pathway enrichment analysis were performed using Metascape data platform,and visualized to construct a ’ disease-pathway-target-component network diagram’.Results:1.Clinical research :The general data of the treatment group and the control group before treatment were comparable(P > 0.05).From the treatment results,the treatment group and the control group were improved in the total score of TCM symptoms,the single symptom score of TCM,the 24-hour dynamic electrocardiogram,heart rate variability and the Lown classification(P <0.05),and the treatment group improved more(P < 0.05).Total score of TCM symptoms : There was no significant difference in TCM symptom scores between the two groups before treatment(P > 0.05),and there was significant difference in TCM symptoms between the two groups after treatment(P < 0.05).After treatment,the effective rate of TCM symptoms was compared between the two groups,the effective rate of the treatment group was 89.3 %,and the effective rate of the control group was 67.9 %.The total effective rate of the treatment group was higher than that of the control group,and the curative effect of the two groups was different(P < 0.05).Single symptom score of traditional Chinese medicine : There was no significant difference in single symptom score of traditional Chinese medicine between the two groups before treatment(P > 0.05),and there was significant difference in single symptom score of traditional Chinese medicine between the two groups after treatment(P < 0.05).24-hour dynamic electrocardiogram : There was no significant difference in the number of ventricular premature contractions in 24-hour dynamic electrocardiogram between the two groups before treatment(P > 0.05).After treatment,the effective rate of the treatment group was 92.9 %,and the effective rate of the control group was 71.4 %.Heart rate variability : There was no significant difference in heart rate variability between the two groups before treatment(P > 0.05).After treatment,both groups had an improvement effect(P < 0.05),and the effect of the treatment group was more obvious(P < 0.05).Lown classification : There was no significant difference in Lown classification between the two groups before treatment(P > 0.05).After treatment,the effective rate of the treatment group was 85.7 %,and the effective rate of the control group was 50.0 %.There was a difference between the two groups(P < 0.05),and the total effective rate of the treatment group was higher than that of the control group.2.The main core active ingredients include 7-Methoxy-2-methylisoflavone(7-methoxy-2-methylisoflavone),Baicalein(baicalein),Diosgenin(diosgen in),Luteolin(luteolin),48 common targets,and the top seven core targets were VEGFA,IL6,IL1 B,TP53,PTGS2,JUN,TNF.Conclusion:1.Wenxin granule can improve the symptoms of patients with ventricular premature beats,good safety,worthy of application.2.Network pharmacology shows that the monarch drug Codonopsis pilosula and the minister drug Polygonatum sibiricum of Wenxin granules may be through7-Methoxy-2-methylisoflavone(7-methoxy-2-methylisoflavone),Baicalein(baicalein),Diosgenin(diosgenin),Luteolin(luteolin)and other components,VEGFA,IL6,IL1 B,TP53,PTGS2,JUN,TNF and other targets.It involves inflammatory signaling pathway IL-17 signaling pathway,hypoxia inducible factor HIF-1 signaling pathway,immune regulation related pathway C-type lectin receptor signaling pathway,PI3K-Akt signaling pathway and other pathways.After blood circulation,metabolism and other biological processes,plasma membrane raft,membrane raft and other components and G protein occasional receptor activity play a role.
