Objective: This study explored the difference of lncRNA in follicular fluids between patients with phlegm-dampness polycystic ovary syndrome and patients underwent assisted reproduction due to male factors through high-throughput sequencing,to deeply understand the pathogenesis and diagnostic markers of phlegm-dampness PCOS and make basis for research lied ahead;Through the analysis of sequencing results,we studied the potential targets of electroacupuncture on phlegm-dampness PCOS,which will help to enrich theoretical researchs on electroacupuncture in PCOS.Method and Content: Experiment 1: Screening the patients with phlegm-dampness PCOS by extracting lncRNA from follicular fluids of two groups through high-throughput sequencing.to have a good command of lncRNA exprssion,with the functional analysis and articles review.Experiment 2: Participants including 70 patients with phlegm-dampness PCOS were divided into experiment group and control group randomly.Electroacupuncture was treated to experimental patients,contrasting the control group’ s patients treated with sham electroacupuncture three times a week until ovulation retrivement,took one month or one menstrual cycle as a course of treatment.By comparing the changes of the integral table after and before different treatments,the days of exogenous application of gonadotropin,the levels of estradiol and progesterone on the day of trigger,the number of occytes obtained,the numbers of fertilization,available embryos and high-quality embryos were all collected.The clinical benifit of electroacupuncture on improving IVF/ICSI in patients with phlegm-dampness PCOS was verified by exploring the potential value of lncRNA.Results: Experiment 1: A total of 20 lncRNAs in the phlegm-dampness type PCOS showed a trend of significant difference compared with control group,covering 17up-regulated and 3 down-regulated lncRNAs enriched in cell metabolism,cell adhesion,etc;The related gene pathway mainly involved in Huntington,Parkinson,splice body and nonalcoholic fatty liver.Experiment 2: Electroacupuncture significantly reduced the number of Gn used,increased the estrogen level on the trigger day,the number of 2PN fertilization and normal fertilization rate(p<0.05);Following the results of the phlegm-dampness syndrome score of PCOS,it was found that the electroacupuncture intervention significantly improved the symptoms(p<0.05),and the changes were significantly different from those of the control group(p<0.05);Based on the lncRNA screened from experiment 1,it showed the level of TPT1-AS1 and PTOV1-AS1 in the experimental group were significantly higher than those of the control group(p<0.05),while the level of PTPRG-AS1 and SNHG14 were significantly much lower(p<0.05).Conclusions:1.Through the high-throughput sequencing,the transcriptomic study of the follicular fluid of patients with phlegm-dampness PCOS was carried out to obtain the differential expression spectrum of lncRNA;2.Electroacupuncture could significantly improve the symptom by increasing the number of 2PN and normal fertilization rate,Electroacupuncture showed high clinical value in PCOS through regulating the expression of lncRNA TPT1-AS1,PTOV1-AS1,PTPRG-AS1,SNHG14.
