Background:Evodia Decoction is a classical compound formula in the Treatise on Typhoid Fever,treating deficiency cold in the liver and stomach and rebellion of turbid yin,and is a representative formula for warming the lid.Clinical studies have shown that Evodia Decoction has good efficacy in depression.Clinical studies have demonstrated the efficacy of Evodia Decoction in depression.Depression is a common clinical psychiatric disorder that is characterized by persistent depressed mood,anxiety with cognitive decline and other clinical symptoms,and has high rates of suicide,self-harm and relapse.Traditional Chinese medicine believes that patients with depression usually have a deficiency of yang qi,yin-cold condensation,and the internal growth of turbid evil leads to the interconnection of phlegm and stagnation,depressing yang qi.Therefore,the basic therapeutic principle is to warm,tonify and descend,and on the basis of keeping in mind the disease mechanism,Evodia Decoction is used to warm the middle and tonify the deficiency,descend the rebellion and stop vomiting,and has a very good healing effect.Purpose:In this study,we investigated the mechanism of antidepressant action of Evodia Decoction,a classical formula from the Treatise on Typhoid Fever,through behavioral and systematic pathological studies.Methods:Animal experiment research was used.Animal experiment:Modeling was performed by CUMS,and the depression-like behaviors of depression model mice were evaluated by sugar-water preference test,water maze test,open field test and forced swimming test;the pathological damage of hippocampus of depression model mice was measured by HE staining;finally,the expression of purinergic receptor P2X,ligand-gated ion channel 7(P2X7R)and NOD-like receptor thermal protein domain associated protein 3(NLRP3)in hippocampal tissue of mice was detected by immunohistochemical staining.Some literature research mainly focuses on the understanding and treatment of depression.the prescription of Evodia Decoction,and the relationship between P2X7R.NLRP3 protein and depression syndrome,etc.Result:Compared with the mice in the model group.Evodia Decoction had a significant ameliorating effect on the depression-like behavior of mice.Evodia Decoction increased body weight,increased sugar-water preference values,shorter immobility time in forced swimming,increased number of platform crossings in the water maze,and increased number of grid walks and uprightness in the open field experiment.Evodia Decoction improved hippocampal pathological damage in mice in a depression model.Further studies showed that Evodia Decoction also reduced the levels of P2X7R and NLRP3,suggesting that its antidepressant mechanism of action may be related to the P2X7R/NLRP3 signaling pathway.Conclusion:Evodia Decoction had better antidepressant effects,improved depression-like behaviors in mice with CUMS-induced depression and attenuated pathological damage in the hippocampus of mice with depression model,For example,improve the loss of pleasure,poor learning and memory,reduced autonomous activities,despair and other conditions.Its mechanism of action may be related to the P2X7R and NLRP3 Proteins in hippocampus.
