Objective:By observing the effects of different retention time of electroacupuncture Tiānshū(ST 25)and ShàngJùxū(ST 37)on intestinal function and serum-related substances in the diarrhea type irritable bowel syndrome(IBS-D)model rats,to investigate the optimal retention time of electroacupuncture in IBS-D model rats.Methods:Sixty SPF Wistar male rats were randomly divided into blank group and model preparation group.The model preparation group included model group,treatment group(the 5 min group,the 10 min group and the 20 min group),with 12 rats in each group.Water avoidance stress method was chosen for modeling,and abdominal retraction reflex method combined with loose stools rate was used for model evaluation.In the treatment group,the rats were treated with electroacupuncture for 5 min,10 min and 20 min,respectively,in the model group,the rats were only grasped and tied,both treatment group and model group were bound for 20 min once a day for 14 days.The blank group was not treated in any way.Behavioral tests were performed on rats before and after modeling and after treatment.Under anesthesia,the ECG signals were recorded under standard II leads for 15 min using the Powerlab data acquisition and analysis system,and then the rat was exposed to the unilateral rectus abdominis muscle,and a pair of platinum wire electrodes were embedded in the rectus abdominis muscle to record the magnitude of colonic pressure at the time of significant contraction of the rectus abdominis muscle stimulated by colorectal dilation.After the measurement,the abdominal cavity of the rats was exposed,and a pair of platinum wire electrodes were embedded in the proximal colonic plasma layer to record the colonic intestinal electrical signals for 2h.The levels of cAMP,cGMP and SP were measured by ELISA.Results:(1)General situation:After electroacupuncture treatment,diet,activity,mental state,excreta and body weight of rats in the treatment group were improved to varying degrees,and the improvement of the 20 min group was better than that of the 5 min group and the 10 min group;(2)Behavioral Science:After electroacupuncture treatment,OT%and OE%in the model group were significantly decreased compared with the blank group(P<0.001),while OT%(P<0.05,P<0.01)and OE%(P<0.05,P<0.001)in the 10 min group and the 20 min group were significantly increased compared with the model group,but there was no statistical difference between treatment groups(P>0.05);(3)Heart rate frequency domain:Compared with blank group,LF and LF/HF in model group were significantly increased(P<0.001),while HF was significantly decreased(P<0.001);Compared with model group,the HF of the 10 min group and the 20 min group was significantly increased(P<0.01,P<0.001),and the LF and LF/HF of the 20 min group were significantly decreased(P<0.001).There was no statistical difference between the 10 min group and the 5 min group(P>0.05),but HF was significantly increased between the 20 min group and the 10 min group(P<0.001);(4)Colonic pain sensitivity:Compared with blank group,colorectal pressure during rectus contraction in model group was significantly increased(P<0.001),while colorectal pressure in treatment group was significantly decreased compared with model group(P<0.001).Compared with the 5 min group,the colorectal pressure in the 10 min group was significantly decreased(P<0.01),and that the 20 min group was significantly decreased(P<0.05);(5)Colonic electromyography:Compared with blank group,the slow wave frequency in model group was significantly decreased,and amplitude of colonic slow was significantly increased(P<0.001);Compared with model group,the slow wave frequency of colon in treatment group was significantly increased(P<0.001),while the amplitude in the 20 min group was significantly decreased(P<0.01);Compared with the 5 min group,the slow wave frequency of colon in the 10 min group was significantly increased(P<0.001),and the slow wave frequency of colon in the 20 min group was significantly increased(P<0.001),and there was no significant difference in colon slow wave amplitude between treatment groups(P>0.05);(6)ELASA serum:Compared with blank group,serum concentrations of cAMP,cGMP and SP in the model group were significantly increased(P<0.001),while serum concentrations of cAMP,cGMP and SP in the treatment group were significantly decreased compared with the model group(SP of the 5 min group:P<0.01;other acupuncture group:P<0.001),compared with the 5 min group,the concentrations of serum cAMP,cGMP and SP in the 10 min group were significantly decreased(P<0.01,P<0.001,P<0.001),and the concentrations of serum cAMP,cGMP and SP in the 20 min group were significantly decreased(P<0.001,P<0.001,P<0.01).Conclusion:1.Electroacupuncture of "Tianshu"(ST 25)and "Shangjuxu"(ST 37)for 5min,10min and 20min can improve the mental state of IBS-D model rats,repair the balance of autonomic nerve and colonic motor function,regulate colonic pain sensitivity and serum related substance levels.2.The regulation of autonomic balance and colic function was better in the IBS-D model rats at density waves of 2Hz and 1mA than at 5min and 10min.It has been tentatively suggested that adequate retention time is required for better efficacy in IBS-D treatment with electroacupuncture.
