Object: At present,the research of MPP2 gene in malignant tumors is increasingly in-depth.Existing studies have found that MPP2 gene is highly expressed in some tumor tissues,but there are few studies in colorectal cancer tissues.Therefore,the specific mechanism of MPP2 gene in the development and progression of colorectal cancer remains unclear.This topic mainly discusses the expression of MPP2 gene in colorectal cancer tissue,and studies the expression of MPP2 gene in colorectal cancer through the analysis of clinical case data and postoperative survival data.The correlation between the expression of MPP2 gene and the clinicopathological features of patients was further analyzed,and the influence of the expression of MPP2 gene on the long-term survival of patients after surgery was also observed.Methods: The postoperative tissue samples of colorectal cancer patients in our hospital were collected,the parts meeting the inclusion criteria were selected,and the expression of MPP2 protein in cancer tissues and adjacent normal colon tissues was detected and analyzed by immunohistochemical staining.The expression of MPP2 protein in different colon tissues was tested and determined by Western blot.Using reverse transcription-polymerase chain reaction(RT-PCR)technique,the m RNA expression of MPP2 in colorectal cancer tissues and adjacent normal colon tissues was detected and compared,and the expression of MPP2 m RNA in colorectal cancer cell lines and normal colon cell lines was analyzed.The influencing factors of different expressions of MPP2 protein in patients with colorectal cancer were analyzed by one-way variance analysis,and the independent influencing factors of prognosis of patients with colon cancer were evaluated by COX regression model.In addition,Kaplan-Meier survival analysis was used to analyze the relationship between different expressions of MPP2 protein and prognosis in patients with colon cancer.Result: The study showed that the expression of MPP2 protein was low in normal colon tissues,but high in colorectal cancer tissues,and the difference was statistically significant.By analyzing the relationship between the expression of MPP2 protein and the clinicopathologic features of colon cancer patients,it was found that the expression intensity of MPP2 protein was also continuously increased when the patients showed peripheral lymph node metastasis,distant invasive metastasis and TNM staging.There was no correlation between the expression intensity of MPP2 protein and the age,sex,primary tumor size and tumor differentiation.The results showed that the expression intensity of MPP2 protein in colon cancer tissues was significantly increased compared with that in normal colon tissues by Western blot assay(P< 0.05).RT-PCR results also confirmed that compared with normal colon tissues adjacent to cancer,the expression of MPP2 m RNA in colon cancer tissues was significantly enhanced,and the difference between the two was statistically significant.It was also found that the expression intensity of MPP2 m RNA in colon cancer cell lines HCT-116 and SW480 was significantly higher than that in normal colon cell lines Fhc,and these conclusions were statistically significant.Kaplan-Meier survival analysis results showed that postoperative colon cancer patients with higher MPP2 expression level had shorter overall survival,and patients with higher MPP2 expression level had significantly lower survival time than patients with low MPP2 expression level,and this difference was statistically significant.In addition,univariate and multivariate COX regression analysis showed that distant invasive metastasis and tumor TNM staging were independent prognostic factors for colon cancer patients.Conclusions: MPP2 protein is highly expressed in colorectal cancer tissues,which can promote the growth and proliferation of colorectal cancer cells.Further analysis showed that the differential expression of MPP2 protein is related to the presence of lymph node metastasis,distant invasive metastasis and tumor TNM staging.It was not related to patient age,sex difference,primary tumor size and tissue differentiation degree.Moreover,the prognosis of colorectal cancer patients with high expression of MPP2 protein is much lower than that of patients with low expression of MPP2 protein.Therefore,MPP2 can be used as a biological indicator of research significance,which can be used to predict postoperative survival of colon cancer patients,and may become a potential target for molecular targeted therapy of colorectal cancer.
