| Objective:Based on the principles of "specificity,effectiveness,and measurability",specific quality control indicators closely related to clinical efficacy and with clear mechanisms were selected for Rhizoma Cyperi.,providing scientific basis for further quality control and quality standard improvement research of Rhizoma Cyperi..Methods: 1.Ultra high liquid chromatography electrostatic field orbital trap high resolution mass spectrometry(UPLC-Q-Orbitrap MS)has been applied to identify the full components of Rhizoma Cyperi.According to the excimer mass,secondary characteristic fragment ions,retention time,Molecular formula and other information,Mass spectral interpretation is carried out to determine the attribution of each chemical component.2.UPLC-Q-Orbitrap-MS is applied to identify the components in Rhizoma Cyperi blood to clarify the specific transitional components in Rhizoma Cyperi blood after administration.3.The components in Rhizoma Cyperi blood are used as target objects for network pharmacology and in vivo models of inflammation and pain to predict and validate targets and pathways,and to screen effective components that play a major regulatory role in each target pathway.4.The testability evaluation and content determination conditions of each medicinal component were optimized,and modern pattern recognition methods were used to evaluate and rank the quality of multiple batches of Rhizoma Cyperig medicinal materials,providing a basis for further quality control and standard improvement research of Rhizoma Cyperi.Results: 1.A total of 43 components were identified from the ethanol extract of Rhizoma Cyperi,including 13 sesquiterpene,4 flavonoids,3organic acids,7 phenols and phenolic acids,6 amino acids,3 coumarins and 7 other types of components,of which 4 components,such as gallic acid,catechin,pyrogallol and hispidulin,were identified for the first time.2.Nine transitional components in the blood of Rhizoma Cyperi.tuberosus were identified by serum medicinal chemistry methods,which were tetradecyl sulfonic acid,pinic acid,artemisinin,pyrogallol,luteolin,ferulic acid,vanillin α-Cyperone and nocarnone are mainly composed of sesquiterpene,flavonoids and organic acids.3.The results of network pharmacology and in vivo model validation indicate that Rhizoma Cyperi.may be mediated by Six pharmacological components such as Cyperenone、aCyperone,act on AKT1 and TNF-α、 IL-6、COX-2、p38MAPK and other targets to regulate p38 MAPK phosphorylation and NF-κB phosphorylation,arachidonic acid and other pathways to achieve the treatment of inflammation and pain and other diseases.4.The testability evaluation results showed that all six components were measurable,and the content determination conditions were optimized.Based on the content results of the six components,grey correlation and TOPSIS methods were used to effectively evaluate the quality of various batches of Rhizoma Cyperi.medicinal materials.Conclusion: This study identified and screened six pharmacological components closely related to the anti-inflammatory and analgesic effects of Rhizoma Cyperi.based on the principles of "specificity,effectiveness,and testability".These components can be used as exclusive property control indicators for Rhizoma Cyperi.,providing a basis for further quality control and standard improvement research on Rhizoma Cyperi.. |
