Rhizoma Chuanxiong-Rhizoma Cyperi herb couple is mainly used for the treatment of depression syndrome, and often used in many traditional Chinese compound prescription. There are significant differences in pharmacological effect when used in different proportions. But the material basis and compatibility mechanism have not been elucidated. In this study, we researched the effect of different proportions and extraction methods on dissolution in vitro of main components, and pharamacokinetic process and tissue distribution of ferulic acid in rat. The results will provide basis for the reasonable combination and clinical application of the couplet.1. Study on the extraction of the Rhizoma Chuanxiong with Rhizoma Cyperi in different ratios.The couplet contains many chemical constituents, water soluble phenolic acids and liposojuble essential oil.. We selected α-cyperone, ferulic acid and ligustilide as index. We established HPLC method for determination of main components to study in vitro dissolution in different couplet ratios by different extraction method. We extracted the couplet by decoction, ultrasonic extraction, petroleum ether and 75% ethanol, and determined the index components by HPLC. The results showed that the dissolution of ferulic acid was slightly higher in water decoction extraction. Ligustilide and α-cyperone as the oil soluble ingredients, the dissolution rate were highest in the petroleum ether and 75% ethanol percolation extraction conditions, and the total dissolution amount of the three components also exhibits a maximum value in this condition.2. Pharmacokinetic study of ferulic acid after orally taken the Rhizoma Chuanxiong-Rhizoma Cyperi herb couple in ratWe established the HPLC-MS/MS method to determine ferulic acid in rat plasma. Rats were given by gavage with the couplet extraction in different ratios. The ferulic acid pharmacokinetic parameters of Cmax, AUC(0-t), AUC(0-∞), MRT(0-t) was significantly higher than other groups(except model) in ratio1∶2(Rhizoma Chuanxiong-Rhizoma Cyperi). The result has significantly statistical difference(P < 0.05). The study suggested that the absorption of ferulic acid could be inproved by increasing the ratio of Rhizoma Cyperi in couplet.3. Study on the tissue distribution of feulic acid in ratRats were administrated with the couplet extraction in different compatibility proportin. We determined the concentration of ferulic acid in heart, liver, spleen, lung, kidney, and brain. Results showed that the distribution of ferulic acid in the liver was higher, but less in the heart and brain. The concentration of ferulic acid were decreased in kidney, liver and brain with the increased propotion of Rhizoma Cyperi in the couplet.We also studied the tissue distribution of ferulid acid in rat of qi stagnation and blood stasis after gavaged with Rhizoma Chuanxiong-Rhizoma Cyperi couplet(1∶2). The results showed that the concentration of ferulic acid increased in blood, heart, brain, lung and spleen in pathological conditions It was of significance to the clinical treatment.
