Objective:To investigate the therapeutic effect of Mongolian medicine Huo Xiao Qi Wei San on mouse calcium oxalate kidney stone model and the influence of osteopontin(OPN),so as to provide scientific basis for clinic。Methods:(1)Before the experiment,the mice adapted to the environment and were fed for 1 week;(2)Seventy KM mice were randomly divided into 6 groups according to body weight:blank group,model group,Potassium Sodium Hydrogen Citrate Granules group,Mongolian medicine Huo Xiao Qi Wei San low-dose,medium-dose and high-dose groups;(3)The blank group was given free drinking water + normal saline 1m L/ day intragastrically,and the other groups were given 1% glycol +1% ammonium chloride 1m L/ day intragastrically for modeling.The modeling lasted for 4 weeks;(4)The weight of mice in each group was recorded every week during modeling,and the mean weight of each group was calculated to observe whether the mice died,ate,behavior and hair changes;(5)After 3 weeks of modeling,one mouse in blank group and model group was sacrificed at random,and HE staining was used to detect whether there were crystals in the kidney of the mouse,and the modeling situation was evaluated;(6)After successful modelling,mice in the model group were given pure water,0.25g/kg in the low-dose Mongolian medicine Huo Xiao Qi Wei San group,0.5g/kg in the medium-dose Mongolian medicine Huo Xiao Qi Wei San group,1.0g/kg in the high-dose Mongolian medicine Huo Xiao Qi Wei San group,1.67g/kg in the Potassium Sodium Hydrogen Citrate Granules group,and the volume of gavage was 20ml/kg.Gavage was given once a day for 3weeks。(7)During the administration period,the weight of mice in each group was recorded weekly,and the mean weight of each group was calculated to observe whether the mice died,feed,behavior and hair changes。(8)Detection index: The content of OPN in kidney tissue was detected by enzyme-linked immunoassay。Results:(1)During modeling,there was statistical difference between the mean weight of mice in the first week of the Mongolian medicine Huo Xiao Qi Wei San high-dose group and those in other groups(P < 0.05),but there was no statistical difference between the mean weight of mice in the second,third and fourth weeks(P > 0.05).;(2)There was no significant difference in mean body weight among all groups during administration(P > 0.05);(3)After 4 weeks of administration,there was statistical significance in OPN between the blank group and the model group(P < 0.05),but there was no statistical significance in OPN between the low-dose group of Huo Xiao Qi Wei San and the model group(P > 0.05),There was statistical significance in OPN between the medium and high dose groups of Huo Xiao Qi Wei San and the model group(P < 0.05);Conclusion:Mongolian medicine Huo Xiao Qi Wei San Powder plays a role in the treatment of calcium oxalate kidney stones through the effects of resolving stagnation and removing stones.Its mechanism is to inhibit the expression of OPN in the kidney,thereby inhibiting the aggregation and nucleation of OPN in the kidney。...
