Objective: To observe the preemptive calm safety and effectiveness of different dose of intranasal dexmedetomidine combined with sevoflurane inhaled anesthesia during hernia sac ligation surgery in children,Exploring the clinically appropriate dosage of dexmedetomidine during pediatric day surgeryMethods: A total of 120 children(108 males and 12 females,aged2 ~ 3,11 ~ 20 Kg weighted,ASA Ⅰ or Ⅱ)scheduled for ambulatory hernia sac ligation surgery were divided into three groups randomly,namely group A,B and C: received intranasal dexmedetomidine 1 ug/kg,2 ug/kg,4 ug/kg in 25~30 minutes before sevoflurane inhaled induction though 2ml syringe.The satisfaction of sedation before the operation,induction time,recovery time body movement rate during the operation,postoperative nausea,vomiting and restlessness were observed.Besides,the satisfaction of families and surgeon were also observed.Results: Compared with the group A,the induction time was shorter,the time of recovery was longer,the body movement,postoperative nausea,vomiting and restlessness were decreased in group B and C(P<0.05).Compared with the group B,the time of recovery was longer in group C(P<0.05).Compared with the group A,the satisfaction rating of sedation before the operation were improved,the same with the satisfaction of families and surgeon.(P<0.05).Conclusion: The sedative and analgesic effect of dexmedetomidine was positively correlated with the dose.Intranasal a DEX inistration of2ug/kg dexmedetomidine is more effective on 2~3 years old children in ambulatory hernia sac ligation surgery.
