Objective:Based on data mining technology,Professor Lv Ying’s medication rules for treating childhood autism spectrum disorder(ASD)were explored,and Li Ke’s academic school theory of traditional Chinese medicine was used to explain the basis of prescription and medication,and then exploring the active ingredients,targets and mechanisms of the core drug group in the treatment of children with ASD through network pharmacology,which can provide theoretical support for the treatment of ASD by traditional Chinese medicine.Methods:1.The medical records of children with ASD treated by Professor Lv Ying in the outpatient department of ancient Chinese medicine of Nanfang Hospital of Southern Medical University from September 2020 to December 2021 were collected.The data of children who met the inclusion criteria were organized into Excel data,and data mining methods such as frequency analysis and association rule analysis were used to summarize Professor Lv Ying’s clinical prescriptions for ASD.2.Selecting the core drug group "Shigao-Wumei" for the treatment of ASD for network pharmacology analysis,and TCMSP、ETCM database and literature were used to collect the active ingredients and targets of traditional Chinese medicine,the disease-related target genes were researched through the Genecards database.Through the Veen diagram to obtain the potential targets of "Shigao-Wumei" in the treatment of ASD,the Cytoscape 3.6.0 software was used to construct a network map to obtain the core targets of the drug group to treat the disease,eventually,the DAVID database was used to conduct GO and KEGG pathway enrichment analysis on the potential targets.Result:1.A total of 103 children with ASD were included in this study,with a total of 262 visits,including 86 males and 17 females,age concentrated in 3-7 years old,including 114 herbal medicines,and the frequency of use of traditional Chinese medicines was 2397.2.Based on frequency statistics,it is found that the most frequently used drugs include Wumei,Shigao,and Chishao ect.High-frequency drug pairs include Shigao-Wumei,Chishao-Guizhi,Chishao-Wumei ect.According to the association rules,the association of Shigao-Wumei was the highest,which was the core drug pair for the clinical treatment of ASD by Professor Lv Ying.3.Network pharmacology analysis screened out the core components such as calcium sulfate dihydrate,Hydrocyanic Acid,beta-sitosterol,Stigmasterol,quercetin from the drug group of "Shigao-Wumei".There are 348 potential targets for the treatment of ASD,of which the core targets are AKT1、ACTB、SRC、HSP90AA1、EGFR、VEGFA、ESR1,etc.GO enrichment analysis results include protein phosphorylation,signal transduction,G-protein coupled receptor signaling pathway and other biological functions,KEGG pathway enrichment analysis mainly involves neuroactive ligand-receptor interaction,PI3K-Akt signaling pathway,Calcium signaling pathway,cAMP signaling pathway,Ras signaling pathway,Rapl signaling pathway and other signaling pathways.Conclusion:1.Professor Lv Ying believes that the general pathogenesis of ASD in children is insufficiency of three yin root Qi,earth fails to suppress fire,meanwhile,the fire,heat,and dryness prevail.In clinical treatment,kidney and spleen are often cared,clear away the fire,heat,and dryness,strengthen the Qi transformation of the san-jiao,etc.Through the fire to generate the earth,the earth to cover the fire,the water to cover the wood,the earth to carry the wood,close Yangming,close Jueyin,and descend the gallbladder to enhance the primordial Qi of source of life and the root of Yang.2.The mechanism of "Shigao-Wumei" treats ASD may be mainly through neuroactive ligand-receptor interaction,PI3K-Akt signaling pathway,Calcium signaling pathway,cAMP signaling pathway,Ras signaling pathway,Rapl signaling pathway and other pathways can improve the clinical symptoms of children with ASD by regulating the liver-brain axis,mediating neuroinflammation,and repairing nerve cell damage. |
