Objective:To observe the effect of bloodletting pretreatment at"Twelve well points of hand"on the expression level of HIF-1α,VEGF of rats with acute hypoxic induced lung injury,so as to investigate the effect of"Twelve well points of hand"bloodletting pretreatment on acute hypoxic induced lung injury.Methods:72 male SD rats were randomly divided into 8 blank control group(hereinafter referred to as control group or N group),32 acute hypoxia model group(hereinafter referred to as model group or D group),and 32 well points bloodletting pretreatment group(hereinafter referred to as bloodletting group or C group).The model group and bloodletting group were divided into 4 subgroups at different time of6h,24h,48h and 72h with 8 rats in each subgroup.The acute lung injury model was established by simulating 15%oxygen concentration(15%O2,85%N2)in the hypoxia simulation chamber of experimental animals.The bloodletting group was pretreated with bloodletting in the order of Shaoshang,Shangyang,Zhongchong,Guanchong,Shaochong and Shaoze,once a day for 5 days.The expression level of HIF-1αin rat lung was detected by Western blot.The expression level of VEGF in rat plasma was detected by ELISA.Results:Morphologically,compared with the control group,the lung tissue structure of the model group was damaged in varying degrees at each time point,indicating that 15%O2acute hypoxic environment can cause acute lung injury,and the injury increased with the extension of hypoxic time.Compared with the model group,the injury of lung tissue in the bloodletting group decreased to some extent at the same time.Compared with the control group,the expression level of HIF-1αand VEGF in the model group increased significantly(P<0.01).Compared with the model group at the same time,the expression level of HIF-1αin the bloodletting group decreased(P<0.05),but the bloodletting group was still higher than the control group at 6h,48h and 72h(P<0.01).Compared with the model group at the same time,the expression level of VEGF decreased at 24h,48h and 72h(P<0.01),but in the each bloodletting group was still higher than in the control group(P<0.05).Conclusions:“Hand twelve well point”bloodletting pretreatment can improve lung injury of the model rats with 15%O2acute hypoxic induced lung injury in a certain time and extent,which may be related to the down regulation of the expression level of HIF-1αand VEGF.
