Effects Of "Jing Acupoint" Bloodletting Pretreatment On AngⅡ,ACE And ET-1 In Rats With Acute High Altitude Lung Injury

Objective: To observe the effect of bloodletting pretreatment at "Twelve well points of hand" on the content of Ang Ⅱ in lung tissue,the content of Ang Ⅱ in plasma,ACE m RNA expression was observed in rat lung tissue and ET-1 content of acute high altitude,provide experimental evidence for clinical application of prevention and treatment of acute high altitude pulmonary injury acupuncture.Method: Male SD rats were divided into groups A(n = 8),hypoxia control group D and well point bloodletting intervention group C.Hypoxia control group D(D6,D24,d48,D72),well point bloodletting intervention group C(C6,C24,C48,c72).Group D and group C were treated with blood sampling needle.The left forelimb was pricked first,and then the right forelimb was pricked.There were 12 acupoints.Selected points: Shaoshang(lung meridian),Shangyang(large intestine meridian),zhongchong(pericardium meridian),Guanchong(Sanjiao meridian),shaochong(heart meridian)and Shaoze(small intestine meridian).The blank group and the hypoxia control group were given the same stimulation way as the well point bloodletting intervention group every day,only grasping without acupuncture.After 5 days,the rats in group C and group D were exposed to simulatedaltitude of 7000 m for corresponding time to prepare the model of high altitude lung injury.Calculation lung wet to dry weight ratio(w / D);he pathological changes in lung tissue was observed;ELISA assay of angiotensin Ⅱ(Ang II)and endothelin-1(ET-1);real-time PCR detection of lung ACE gene expression.result:1.Lung W / D: Group D W / D in comparison with A group,the time period W /D significantly increased,and the value changed with time,reaching the peak at 48 h,indicating that low-pressure hypoxia can increase w / D in lung tissue.2.He results: compared with group A,pathological sections of group D were damaged in different degrees,and the damage was the most serious at 72 h.The results showed that hypobaric hypoxia could induce lung injury in rats,which was aggravated with the time of hypoxia.At the same time,the lung tissue injury of acupuncture groups was improved in varying degrees compared with model groups.3.Compared with the control group A,group D ANG II and ET-1 levels increased significantly at each time period,as prolonged hypoxia and gradually increased,suggesting that low-pressure hypoxia can increase the levels of ANG II and ET-1 in lung;At the same time,the content of Ang II and the content of ET-1 in lung tissue in acupuncture groups were decreased compared with those in model groups.4.Real time PCR results: compared with group A,the expression of acemrna in group D was up-regulated at all time points,suggesting that low-pressure hypoxia can up regulate the expression of acemrna in lung tissue.Except for 72 hours,At different time points after the intervention of acupuncture,and the remaining three groups of ACE m RNA expression were decreased.Conclusion: 1.through this experiment,it is verified that the artificial low pressure oxygen cabin equipment can induce lung injury in normal rats.2.the results show that the model of lung injury can be established in the model of 7000 meters and72 hours exposure.3.Well point bloodletting pretreatment can affect ang Ⅱ content,ET-1 content and ACE m RNA expression of rats,and then play a preventive and therapeutic role on acute high altitude hypoxic lung injury in rats,providing experimental basis for acupuncture prevention and treatment of acute high altitude hypoxic lung injury.
