Objective: To evaluate the association between NF-κB and age-related macular degeneration(AMD).Then observe the therapeutic effect of Ziyin Mingmu Decoction on the rat model of AMD and explore its possible internal mechanism.Methods: Clinical study: 55 patients with Age-related macular degeneration treated in the ophthalmology department of Qinghai Provincial People’s hospital were selected as the case group,and 57 healthy people who underwent physical examination in the physical examination center of Qinghai Provincial People’s Hospital in the same period were selected as the control group,draw peripheral blood,sequencing of single nucleotide polymorphisms at four loci of NF-κB gene rs3774959,rs3774932,rs3774937 and rs230526.Experimental study: 50 SD rats were randomly divided into five groups,except for the blank group,the remaining groups used self-made modeling devices to model Age-related macular degeneration,of which 10 rats were used for model evaluation.After confirming the successful completion of modeling,Ziyin Mingmu Decoction and corresponding control drug(vitamin E nicotinate capsule)were continuously administered by gavage for 15 days.Then the retinal tissue of rats was taken for HE to observe the pathological changes,and then q RT-PCR,immunohistochemistry and Western blot were used to detect the expression of NF-κB in rat retina.Results:1.The frequencies of AA genotype of rs3774932 and CC genotype of rs3774937 in the case group were significantly higher than those in the control group(P < 0.05);2.Retinal light injury can lead to pathological changes in rat retinal tissue;3.After the intervention of Ziyin Mingmu decoction,the histopathological changes of rat retina were significantly improved;4.4.Ziyin Mingmu decoction can significantly reduce NF after intervention-κ B expression,play its anti-inflammatory role,so as to protect retinal tissue.Conclusion: AA type of NF-κB gene rs3774932 and CC type of rs3774937 may be risk factors for AMD.Ziyin Mingmu decoction can significantly improve the pathological changes of retinal tissue structure in rats.And by regulating NF-κ B gene and protein expression to reduce the level of retinal inflammation,so as to play a role in retinal protection. |
