Background: Surgical treatment is preferred for early hepatocellular carcinoma,while for unresectable hepatocellular carcinoma,including patients with insufficient liver reserve,lenvatinib has been the first-line treatment.In phase III clinical trials,lenvatinib has a more precise effect than sorafenib.Currently,the effect of preoperative targeted therapy with lenvatinib on liver regeneration is not clear.Objective: The effect of lenvatinib on liver regeneration is still unclear.The present study attempted to investigate the effect of lenvatinib on liver regeneration and related molecular mechanisms.Methods: We useed lenvatinib two weeks before hepatectomy and construct the mouse hepatectomy model.By detecting serum samples and tissue samples at different time points.And testing the expression levels of cell proliferation protein and inflammatory factors by Western blotting,Also tested the expression and mechanism of proliferation gene after hepatectomy by real-time quantitative PCR.Results: We found that compared with sorafenib,preoperative application of lenvatinib had the same inhibitory effect on liver function,but less inhibitory effect on liver regeneration,and even promoted cyclin-D1 expression could be observed at the gene level one day after hepatectomy.Conclusion: Nether cut for his role in the process of liver regeneration IL-6 gene and the interference between the TNF alpha channel is small,the side reflect the mechanism could involve drugs affect liver regeneration to the nucleus gene expression or extracellular signal factor one of them is given priority to,and shimron cut for the inhibitory effect of liver regeneration is smaller,if the new auxiliary were used in the treatment of liver cancer targeted therapy,Lenvatinib would be a better choice. |
