| Berberis amurensis Rupr is a plant belonging to Berberis family.Its roots and branches mainly contain alkaloids,which have good pharmacological activities in anti-inflammatory,antibacterial and anti-tumor aspects.In order to ensure the quality of berberis and develop the medicinal value of berberis,in the formation of the quality standard of Chinese medicine berberis in Jilin Province,also reasonable evaluation of the quality of berberis macrophylla.In this paper,the quality evaluation of berberis from different origin,the same origin with different years and different growth modes(cultivation,wild)was carried out.The main contents include:characters,identification items,examination items,extract,determination of three alkaloids and HPLC characteristic chromatogram.The conclusions are as follows:1.A study on the characters and identification of roots and branches of BerberisThe root and branch characters,microscopic and thin layer identification of different batches of berberis macrophylla were studied by visual observation,microscopic imaging and thin layer chromatography.Character: berberis root is kind cylindrical,have branch,skin gray brown,have fine wrinkle,easy to peel off.Hard,not easy to break,uneven section,bright yellow,pith brownish yellow;Branches gray yellow,ribbed,without warts,internodes have sharp thorns,sparse single.Microscopic identification: berberis powder was yellow or yellowish brown,single or bundle of pericardium fiber,obvious pores and grooves,stone cells were yellowish brown,calcium oxalate square crystal;The branch powder was light yellow,the middle sheath fiber was more and fine,the conduit was perforated conduit.TLC identification: The optimum extraction method was used to prepare the test solution,berberine hydrochloride was used as the reference substance,n-butanol: water: acetic acid(2:1:0.5)was used as the developing agent.In the test samples chromatogram of 16 batches,fluorescent spots of the same color appear at the corresponding position of the control sample.TLC identification of different manufacturers,conditions have no influence on TLC identification,all meet the requirements,proving that TLC identification durability is good.2.Study on root and branch examination items and extract of BerberisThe contents of moisture,total ash and heavy metals in root and branch of berberis were determined by drying method,ash determination method and atomic absorption spectrophotometry.The results appear that the moisture content of berberis root was between7.10% and 11.03%,with an average of 9.05%.The moisture content of branches was between7.23% and 9.37%,with an average of 8.46%.The proposed water content of roots should not be over 12.0% and branches should not be over 11.0%.The total ash content of berberis is mostly between 0.49% and 1.93%,with an average of 1.38%,and branches between 0.71% and 2.32%,with an average of 1.32%.After analysis,the total ash content of berberis macrophyllum and branches shall not exceed 3.0%.The results of heavy metal determination showed that the content of samples did not exceed the limit range.Using 50% ethanol as solvent,hot leaching method to measure the extract content of samples,the results appear that the extract content of berberis in 8.01%-19.79%,branches in 7.76-18.32%,considering the origin,different growth and annual growth,through data analysis,to specify the extract of berberis and branches shall not be less than 9.0%.3.Determination of three alkaloids in roots and branches of BerberisA HPLC method for the determination of jatrorrhizine hydrochloride,palmatine hydrochloride and berberine hydrochloride in different batches of berberis root and branches was established.The data appear that the content of berberine hydrochloride in the roots and branches was higher,followed by jatrorrhizine hydrochloride and palmatine hydrochloride.The content of alkaloids in the cultivated berberis is higher than that in the wild.The content of alkaloids in the 2-year old and 9-year old berberis is higher than that in the wild berberis in Weishahe town of Linjiang city and Tonghua city.Linjiang city reed east and dunhua blackstone alkaloid content is higher in amur barberry root branches.It is proposed that the content of berberine hydrochloride should be taken as the index of its content determination,and the content of berberine hydrochloride in the root of berberine not less than 0.6%,branches not less than 0.1%.Different column temperature,three flow rates and the proportion of mobile phase,different HPLC and chromatographic conditions had little effect on the contents of three alkaloids,which all met the requirements,proving that the method had good durability.4.Establishment of Characteristic spectrum of BerberisThe establishment of HPLC method for the determination of rhizobine hydrochloride,palmatine hydrochloride and berberine hydrochloride in the root of Berberis,application of analysis software,draw 16 different batches of amur barberry root root HPLC feature maps,a total of seven common peak is determined,and testify against the three peaks(medicine root alkali,hydrochloric acid berberine hydrochloric acid,hydrochloric acid and Martin).The similarity was over 0.95,conform to the provisions.The HPLC characteristic chromatogram of Berberis macrophylla showed good stability,which could provide basis for the qualitative identification of berberis,and could be used for the quality evaluation of Berberis. |
