This paper studied on pharmacognosy identification, the total alkaloid content of different parts,antibacterial in vitro, anti-tumor activity in vivo and other aspects of Berberis amurensis Rupr.and according to the pharmaclolgical activities, this paper had a preliminary stuty on preparation process、total alkaloid content and antibacterial in vitro of the tincture with the root of Berberis amurensis Rupr. to treat oral ulcer.The research of Berberis amurensis Rupr.was less,and the relebant literatute were also less,for this reason,this text summaried the essential components and the pharmacological activities of the plants of berberis for the experiments reference.This paper summaried15kinds of protoberberine alkaloids,24kinds of bisbenzylisoguinoline alkaloids,47kinds of other alkaloids,and summaried main pharmaclolgical activities of berberis plants,as diminish inflammation,antibacterial activities,reduce blood press and blood glucose,anti-arrhythmia and anticancer,and so on.On pharmacognosy,studied the origin of Berberis amurensis Rupr.and the characteristics of its root.Used the optical microscope to observe powder characteristics and transverse section characteristics of root and stem of Berberis amurensis Rupr.and.studied their internal structure.Powder microscopy observations show that there were many cork cells,wood fibers,conduits,bast fibers and parenchyma cells in the root and stem of Berberis amurensis Rupr.The stone cells less,and more of them were scattered,slender fibrous stone cells;found a small amount of calcium oxalate crystal in the fiber bundles,mostly of them were diamond-shaped and irregular shape,they were difficult to observe;there had calcium oxalate crystal in parenchyma cells,too;the wood fibers into a bundle existed,and they wall were thin. Transverse section microscopy observations show that the adventitial cells of the fibrous root of Berberis amurensis Rupr.were obviously,moer of them were rectangle cells,and aligning close up,the shape of its xylem was star angular and arranged in four beams extending to the vascular cylinder center,so the fibrous root had no pith,there were yellow-green phloem between every two beams of xylem,the cells spreaded close up;and the epidermis of the stem were yellow,most of them were deciduous.Most of the dork layer cells were like square or anomalous shapes,and arranged close up.Cells wall were thickend.Cork cortex cells were less, like square,arranged close up.The cortex connected with the bast,the fiber bundles of the bast were evident.Cambium ring was also evident,and connected with the xylem.The ray of the xylem were wider,has many catheters and fiber bundles.The pith rays were reached the pith, they shape were like horn and composed with1-2tiers cells.The pith was evident,the cells were like round and arranged close up.This experiment used the ultraviolet-spectrophotometer setting-out total alkaloid content of roots, stems, leaves of Berberis amurensis Rupr. And the results show that total alkaloid content of each place of this plant from high to low was the root> stem> leaf. This experiment also contrasted the total alkaloid content of Rhizoma Coptidis. Cortex Phellodendri and Berberis amurensis Rupr.The result was Rhizoma Coptidis>Cortex Phellodendri Amurensis>Berberis amurensis.The experiment of antibacterial in vitro used the paper disk method,studied on the inhibition effect of75%ethanol extract、the total alkaloid、berberine on E.coli、S.aureus、P.aeruginosa、S.entria.The result displaied that all the extracts had better inhibition effect on all tested strains. When increased the concentration of the extracts that the inhibition efficiency would be upgrade.So when the75%ethanol extract and the total alkaloid in concentration for100mg/mL,50mg/mL and25mg/mL that the inhibition effect on P.aeruginosa were best,the inhibition rate were93.73%、89.03%、54.87%、93.23%、64.19%、55.28%.When the berberine in concentration for2.5mg/mL and1.25mg/mL that the inhibition effect on P.aeruginosa were best,the inhibition rate were6133%、55.03%.This experiment studied on the anti-tumor action of75%ethanol extract and the total alkaloid on H22rumor of mice.The result displaied that all the extracts had better anti-tumor action. When the75%ethanol extract and the total alkaloid in concentration for500mg/kg,their anti-tumor efficiency were similar,greater than60%.When their concentration reached1000mg/kg,they had same anti-tumor efficiency and the result was the best, could achieve73.83%;the medication administration teams compared with the negative control groups that the disparation was very significant(P<0.01).Two extracts also had effect on H22tumor of mice. Negative group index of immune organs and the blank group contrast showed significant difference(P<0.01),so the immune oragans of mice would be affected due to tumor-bearing.When75%ethanol extract and the total alkaloid in high and low dose groups, the thymus index were higher than the CTX group,and lower than the negative control group. The spleens index were higher than the CTX group,two tested samples at a dose of1000mg/kg, the spleens index were lower than the negative control group,and closed to the CTX group,so when the75%ethanol extract and the total alkaloid in concentration for1000mg/kg would effect on the immune organs of tumor-bearing mice; the spleens index showed the medication administration teams compared with the CTX group and the normal group the disparation were very significant(P<0.01).So this experiment showed that75%ethanol extract and the total alkaloid could accommodate immunocompetence on H22tumor of mice.This paper also manufactured the tincture of Berberis amurensis Rupr.to treat the oral ulcer,its characteristics and the color were stabile. And set out the total alkaloid content and bacteriostatic activity,and the results were patency.So this paper provided a basis for development of the product in the future.
