| Objective:Through detecting the expression of nuclear pore membrane protein POM121 in hepatocellular carcinoma(HCC),we aimed to analyze the relationship between the expression of POM121 and the clinicopathological characteristics of HCC patients.Then we studied the effect of POM121 on the biological function of HCC cells through in vitro experiments,and explored the mechanism and prognostic significance of POM121 in HCC immune environment.Methods:The expression of POM121 in 75 cases of HCC and its corresponding adjacent non-tumor tissues were detected and analyzed by tissue microarray and immunohistochemistry.After the overexpressed POM121 HCCcell line was constructed,the biological functional changes of the cells were detected and observed by CCK8,Transwell,plate cloning and cell scratch test.The correlations between POM121 withimmune cells and immune regulatory factors were determined by bioinformatics analyses.Apolygenic risk prediction model was developed a prognostic nomogram with calibration curve was constructed to predict the 3-year and 5-year survival probability of individuals.Results:The positive rate of POM121 in HCC tissues was 64.00%(48/75),while the positive rate in adjacent tissues was 42.67%(32/75),and the difference was statistically significant(P<0.05).The expression of POM121 was related to liver cirrhosis and distant metastasis of HCC patients(P<0.05).Furthermore,the survival time of HCC patients with POM121 positive expression was significantly shorter than those with POM121 negative expression,suggesting that POM121 positive expression may be related to the poor prognosis of HCC patients.It was found that the proliferation,invasion and migration of HCC cells were significantly enhanced by over-expressing POM121.Through bioinformatics analysis,POM121 expression was found positively correlated with T help cells(THC),central memory T cells(TCM),and negatively correlated with plasma cell like dendritic cells(PDC),cytotoxic cells and dendritic cells.We identified the association between POM121 with 1,515 immunosuppressive factors and 31 immuno-stimulatory factors.Combined with relevant immunomodulators,a personalized nomogram for predicting the prognosis of POM121 is constructed,and its C index is 0.7209,which can better predict the prognosis of HCC patients.Conclusion:(1)POM121is highly expressed in HCC tissues and may be associated with poor prognosis of HCC patients.(2)POM121overexpression can promote the proliferation,migration and invasion of HCC cells.(3)POM121may be a potential immune prognostic marker. |
