Objective: By probing bioinformatics database,the expressions,possible functions and signal pathways of AIM2 in thyroid carcinoma were preliminarily explored,and then immunohistochemical method was used to study the expression of AIM2 inflammasome in papillary thyroid carcinoma and its relationship with other clinical parameters.The purpose of this study is to explore whether AIM2 inflammasome may become a detection index in the diagnosis and treatment of thyroid carcinoma in the future,or make a preliminary exploration for immunomodulatory therapy and targeted therapy of thyroid carcinoma.Methods: 1.Download the data of thyroid carcinoma in TCGA database,analyze the expression differences of AIM2 between thyroid carcinoma tissues and paracancerous thyroid tissues,and the data were grouped according to the basic information and clinical information of patients,and the differences of AIM2 expression in each group were analyzed respectively.Next,the Kaplan-Meier Plotter was used to analyze the correlation between AIM2 expression and OS in thyroid carcinoma patients.2.Through GO enrichment analysis and KEGG enrichment analysis,the biological processes and signal pathways that AIM2 might participate in thyroid carcinoma were predicted.3.Collect the tissue samples diagnosed as papillary thyroid carcinoma from the Department of Pathology,Affiliated Hospital of Guilin Medical University,and make paraffin sections,take the paracancerous thyroid tissues as the control,and carry out immunohistochemical experiments on the paraffin sections.Chi-square test was used to analyze the differences of the expression of AIM2 inflammasome in papillary thyroid carcinoma and paracancerous thyroid tissues,and the correlation with related clinical indicators.Results: According to the findings of a study on thyroid carcinoma downloaded from TCGA database,the expression of AIM2 in thyroid carcinoma and paracancerous thyroid tissues was different,and there was a statistically significant difference(P<0.05);AIM2 expression was associated to race,the expression level of AIM2 in Caucasian thyroid cancer patients was higher than African-Americans,and there was a statistically significant difference(P<0.05);AIM2 expression was associated to the pathological classification of thyroid carcinoma,the expression of AIM2 in classical thyroid papillary carcinoma was higher than follicular papillary thyroid carcinoma(P<0.01).AIM2 expression in thyroid carcinoma patients with different gender,age and lymph node metastasis was not statistically significant(P>0.05).The expression degree of AIM2 was not longer related to the overall survival of thyroid carcinoma patients(P>0.05).The results of GO enrichment analysis show that the Biological Processes of AIM2 enrichment in thyroid carcinoma include immune response,inflammatory response,apoptotic process,positive regulation of interleukin-1β secretion and so on.KEGG signal pathway analysis shows that the signal pathways of AIM2 in thyroid carcinoma mainly include NOD-like receptor signaling pathway and Cytosolic DNA-sensing pathway.Immunohistochemical experiment showed that the expression of AIM2 was positive in cytoplasm and cell membrane of papillary thyroid carcinoma,while the expression was negative in paracancerous thyroid tissues.The expression of AIM2 in papillary thyroid carcinoma is related to the tumor size(P<0.05),but no related to lymph node metastasis,different clinical stages,different ages and different genders(P>0.05).Conclusion: 1.AIM2 expression in papillary thyroid carcinoma is higher than that in paracancerous thyroid tissues,and related to the size of the tumor,but not related to lymph node metastasis,age,sex and overall survival. |
