Objective By analyzing and comparing the expression of NLRP3 and AIM2inflammasome-related genes in the peripheral blood of patients with different disease stages of brucellosis and the expression of inflammatory cytokines IL-18,IL-1β,IFN-γ,analyzing the correlation between the two and discussing the possible mechanism of action of NLRP3 and AIM2 inflammasomes at different stages of brucellosis provide clues for studying the immune and escape mechanisms of brucellosis.Methods:A total of 40 patients diagnosed with brucellosis in the Affiliated Hospital of Inner Mongolia Medical University and Inner Mongolia Centers for Disease Control and Prevention from May 2020 to January 2021 were collected,including 20 in the acute phase and 20 in the chronic phase.The information was collected and entered into the database。At the same time,20 healthy controls were collected from the Physical Examination Center of the Affiliated Hospital of Inner Mongolia Medical University。All participants in the group collected 2ml of cubital venous blood on an empty stomach in the early morning.Quantitative real-time PCR and ELISA kits were used to detect the expression of NLRP3 and AIM2inflammasome-related genes in the peripheral blood of the diseased and healthy members and the peripheral blood of each group The protein expression levels of inflammatory cytokines IL-18,IL-1β,IFNγ,and the correlation between NLRP3,AIM2 inflammasomes and inflammatory cytokines IL-18,IL-1β,IFNγ.Results:The gene expression level of NLRP3 receptor is between the brucellosis acute phase group(0.78(0.63,1.37)),chronic phase group(0.71(0.39,1.77))and healthy control group(1.00(1.00,1.00))There is no significant difference in the expression of(P>0.05)。The gene expression of AIM2 receptor in the acute stage of brucellosis group(2.17(1.15,3.87))was significantly higher than that in the healthy control group(1.00(1.00,1.01)),and in the chronic stage group(0.63(0.29,1.46)))Was significantly lower than the healthy control(1.00(1.00,1.01)),the difference was statistically significant(P<0.05);The gene expression level of ASC in the acute brucellosis group was 0.85(0.53,0.96)lower than that of the healthy control group(0.80(0.56,1.01)),the difference was statistically significant(P<0.05);The gene expression of Caspase-1 in the chronic stage of brucellosis group(1.53(1.07,2.47))was higher than that of the healthy control group(1.00(1.00,1.01)),and the difference was statistically significant(P< 0.05);IL-18 protein expression in the acute phase group was higher than in the chronic phase,and at the same time higher than the healthy group,the difference was statistically significant(P<0.01);IL-1β protein expression in the acute phase group of brucellosis There is no significant difference in the expression between the chronic phase group and the healthy control group(P>0.05);the expression of IFN-γ protein in the brucellosis acute phase group and the chronic phase group is higher than that of the healthy control group,and the difference is statistically significant Academic significance(P<0.01).Conclusion:1.The gene expression of NLRP3 receptor has no significant difference between the brucellosis acute stage group,the chronic stage group and the healthy control group,and there is a negative correlation with the expression of IL-18.It is speculated that the NLRP3 receptor has an effect on Brucellosis.The sensitivity of the bacteria is not high.In brucellosis,the NLRP3 inflammasome may be inhibited in some way.2.The gene expression of AIM2 receptor in the acute stage of brucellosis was significantly higher than that of the healthy control group,and in the chronic stage it was significantly lower than that of the healthy control group,and it was positively correlated with the expression of IL-18 and IFN-γ protein.It is inferred that AIM2 The inflammasome may play an important role in the body’s defense against brucella infection in the early stage,but may be suppressed in the chronic phase of the disease3.The gene expression level of ASC in the acute stage of brucellosis is lower than that of the healthy control group.It is speculated that the expression of ASC,an important component of the inflammasome in the early stage of brucellosis,is inhibited,which may be the reason that Brucella escapes immunity A way of systematic clearance.4.The gene expression level of Caspase-1 in the chronic phase of brucellosis is higher than that of the healthy control group.It is speculated that it may play a major role in the chronic phase of the disease.
