Objective: To explore the biological function and clinical significance of CDC20 in bladder cancer by TCGA database,bioinformatics and immunohistochemistry methods.Methods: Bladder cancer m RNA data and corresponding clinical data were downloaded from the cancer genome map(TCGA)database.Bioinformatics analysis was used to compare the difference of CDC20 m RNA in bladder cancer and adjacent normal bladder tissues.The TIMER database was used to verify the results.Combined with the relevant clinical data in TCGA database,the correlation between CDC20 m RNA expression level and clinical features,diagnostic value and prognosis were analyzed,and the pathway correlation was analyzed by GSEA analysis method.Immunohistochemical staining was performed on 34 cases of bladder cancer and 11 cases of normal Formalin fixed paraffin embedded tissue(FFPE).The expression level of CDC20 protein was detected by immunohistochemistry,and the correlation and diagnostic value of CDC20 protein with clinical characteristics and prognosis were analyzed.HPA database was used to verify the expression level of CDC20 protein.Results:The differential expression analysis and paired differential analysis of TCGA database and timer database showed that the CDC20 m RNA level in bladder cancer tissue was significantly higher than that in normal bladder tissue adjacent to the cancer(p=2.827e-10;p=1.181e-07).Clinical correlation analysis showed that the expression of Cdc20 m RNA in high-grade bladder cancer was higher than that in low-grade bladder cancer(p=1.9e-07).Prognosis analysis showed that patients with high expression of CDC20 had shorter overall survival time and worse prognosis(P < 0.05).The immunohistochemical results of FFPE cohort and HPA database showed that the positive expression rate of CDC20 protein in bladder cancer tissues was higher than that in normal bladder tissues(p<0.05).The positive expression of CDC20 was related to pathological grade.The subject working curve analysis showed that CDC20 m RNA and CDC20 protein had good diagnostic value in the diagnosis of bladder cancer(area under the curve was 0.928,sensitivity 0.789,specificity 0.860;area under the curve was0.715,sensitivity 0.636,specificity 0.794).GSEA analysis showed that genes involved in cell cycle,bladder cancer,p53 signaling pathway,ubiquitin mediated proteolysis,erb B signaling pathway,MAPK signaling pathway and other related genes were significantly enriched in CDC20 overexpression group,and were involved in arachidonic acid metabolism,cytochrome P450 metabolism of heterologous substances α Genes related to linolenic acid metabolism and glycerol phospholipid metabolism were enriched in the low expression group(P <0.05,FDR < 0.25).Conclusion: CDC20 is highly expressed in bladder cancer tissues,which can be used as a potential biomarker for the diagnosis and prognosis of bladder cancer.The development of drugs targeting CDC20 has potential benefits for the treatment of bladder cancer.
