| Objective:Bladder cancer was one of the most common malignant t umor in male genitourinary system, metastasis was the main cause of deat h for advanced bladder cancer. From researchs about metastasis of bladder cancer, it had been found Rho GDP dissociation inhibitor 2 (RhoGDI2) can inhibited the metastasis of bladder cancer, RhoGDI2 expression in bla dder cancer cells with high metastatic potential were significantly lower th an the low metastatic potential cells. But whether RhoGDI2 was associate d with the invasion and metastasis of bladder cancer was unkowned, and its specific mechanism was not clear. Therefore, our study research RhoG DI2 expression in bladder cancer, made it clear whether RhoGDI2 can in hibited metastasis of bladder cancer at the level of protein, and further an alysis its mechanism, mainly for the signaling pathways and downstream and upstream factors.Method:We researched RhoGDI2 expression in bladder cancer to ma ke it clear whether RhoGDI2 was associated with the invasion and metast asis of bladder cancer. Collected 44 cases of bladder cancer tissues from Affiliated Tumor Hospital of Guangxi Medical University between October 2010 to June 2014. Detected RhoGDI2 expression for its localization and quantitative using immunohistochemistry method. Divided the samples de pend on different pathological parameters and compared between them to see whether there was statistically significant difference. P<0.05 meaned t here was statistically significant difference.Used bioinformatics to examine the genes and signaling pathways ass ociated with RhoGDI2 that inhibited metastasis of bladder cancer,and exp lored the upstream and downstream factors in signaling pathways.Result:All the bladder cancer samples had RhoGDI2 expression, there was no statistically significant difference (P<0.05) between different gender and age groups. For different stages, no statistically significant difference between T3 and T4 stage, beside that,the RhoGDI2 expression was correlated with the stages. For different degrees of differentiationã€primary and recurrentã€metastasis or not groups, there was statistically significant difference (P<0.05).Six series of microarray expression profiles of bladder cancer were downloaded form GEO datasets, we found that A2M,CORO1A,ENC1 and FGD34 were highly overlapping differentially expressed genes, and there were 14 highly overlapping signaling pathways that up-regulate differentially expressed genes enriched, include leukocyte transendothelial migration pathway and so on.Conclusion:RhoGDI2 inhibited the invasion and metastasis of bladder cancer on protein expression level, it may be metastasis suppressor factor for bladder cancer.RhoGDI2 may regulated invasion and metastasis of bladder cancer vi a Vav-RhoA-ROCK-MLC and Ras-Raf-MEK. The downstream factors of t he signaling pathways may be ROCK, mDia1,PAK,MEK, AP1 and MLC; Upstream factors may be Vav, FAK, Src, P13K, PIP3, etc, among them, P AK, AP1, P13K and PIP3 had never been reported. |
