Objective: To detect the effects of Jinkui Shenqi pills combined with Buzhong Yiqi pills on exercise capacity,body weight,body temperature of different parts,blood glucose in old mice;to detect the effects of Jinkui Shenqi pills combined with Buzhong Yiqi pills on the protein expressions of AMPKαand P-AMPKα in the internal organs of old mice,to preliminarily explore the regulatory mechanism of Jinkui Shenqi Pills combined with Buzhong Yiqi Pills on key molecules of bioenergy metabolism,and provide theoretical and experimental basis for the prevention and treatment of of aging-induced decline in physiological and metabolic functions and related diseases.Methods: Natural aging mouse models(18-24 months old)were built and randomly divided into normal control group(Contorl),Jinkui Shenqi pills group(JKSQ),Buzhong Yiqi pills group(BZYQ),Jinkui Shenqi pills combined with Buzhong Yiqi pills group(JKSQ & BZYQ)according to the similar body weight.Tracing the body weight,body temperature of different body parts and blood glucose of old mice,evaluated the exercise capacity of old mice by balance bar test,open field test and suspension test,to detect the physiological regulation of Jinkui Shenqi pills combined with Buzhong Yiqi pills in old mic.The protein expressions of AMPKα and P-AMPKα in liver,pancreas,kidney,spleen,jejunum and colon of old mice were detected by Western blot(WB)and immunofluorescence analysis.Results: 1.The body weight data showed that compared with before administration,the body weight of old mice in the Contorl group and the BZYQ group decreased significantly after administration(P<0.05),while the body weight of JKSQ & BZYQ group did not change significantly.2.The body temperature data of different parts showed that the head,neck,chest and abdomen of old mice which close to the heart had higher temperature,the limbs and tail had lower temperature,especially the left lower limb and tail had the lowest temperature,it showed that the body temperature had a centripetal distribution.After administration,the JKSQ & BZYQ group old mice increased the body temperature,the temperature of right upper limb,left lower limb and right lower limb were significantly higher than those in the Contorl group(P<0.05).Compared with before administration,the temperature of left lower limb and right lower limb of old mice in the JKSQ & BZYQ group also were increased significantly(P<0.05).3.The fasting blood glucose data showed that compared with before administration,the blood glucose of old mice in the Contorl group increased significantly(P<0.05).After administration,the blood glucose level of old mice in the JKSQ & BZYQ group was closer to the normal value than that in the Contorl group(P<0.05).4.Behavioral data showed that,(1)In balance bar test: compared with before administration,the time spent on balance bar of old mice in the Contorl group and the BZYQ group increased significantly(P<0.05),while JKSQ & BZYQ group decreased the time spent on balance bar,but there was no significant difference.(2)In open field test:compared with before administration,old mice in the Control group,JKSQ group and BZYQ group decreased the open field movement distance,while the JKSQ & BZYQ group increased the distance,but there were no significant differences.(3)In suspension test: compared with before administration,the time spent in suspension of old mice in the Control group was significantly reduced(P<0.01).After administration,the JKSQ group,BZYQ group and JKSQ & BZYQ group were significantly increased the suspension time than that in the Control group(P<0.001).5.Western Blot results showed that AMPKαand P-AMPKα proteins were expressed in the liver,spleen,kidney,pancreas and colon tissues of old mice,and the expressions of AMPKα and P-AMPKα of old mice in the JKSQ & BZYQ group were significantly higher than those in the Control group(P<0.05).6.The immunofluorescence analysis showed that The expressions of AMPKα and P-AMPKα in the liver,spleen,kidney,pancreas and colon of old mice in the JKSQ & BZYQ group were significantly increased than those in the Control group(P<0.05).Conclusions:(1)The combination of Jinkui Shenqi Pills and Buzhong Yiqi Pills can regulate the body weight,body temperature,blood glucose and exercise capacity of old mice.(2)The combination of Jinkui Shenqi Pills and Buzhong Yiqi Pills may increase the activity of AMPKα in the tissue of old mice,activate the AMPK pathway,and improve the decline of physiological and metabolic functions caused by aging. |
