| Background:Nasopharyngeal carcinoma(NPC)is an EB virus-associated malignant tumor.Because of its hidden incidence site,some patients are in advanced diseases when initial diagnosed,and have poor treatment outcomes.Small RNA(micro RNA,miR)is a short-chain RNA with a non-coding protein that regulates target gene expression at the posttranscriptional level,and its expression imbalance is closely related to the development of multiple human diseases,including NPC.Our previous small samples study found that the expression of EBV-miR-Bart1-3p and hsa-miR-23b-3p in NPC tissues of Guangxi was significantly higher than that in nasopharyngeal mucosal chronic inflammatory tissues.How their expression and clinical significance in NPC tissues need to be further studied.Objective: To study the expression of EBV-miR-Bart1-3p and hsa-miR-23b-3p in NPC tissues of Guangxi,and analyze the differential expression level between the NPC tissuses and nasopharyngeal mucosal chronic inflammatory tissues,as well as the correlation between the expression level in NPC tissues and the patients’ clinicopathological characteristics and Disease-free survival time.Try to discover new NPC oncogene or tumor suppressor genes,and provide theoretical basis for NPC targeted therapy and further study of its pathogenic mechanism.Methods: The expression of EBV-miR-Bart1-3p and hsa-miR-23b-3p were detected by real-time PCR(Quantitative Real-time PCR,q RT-PCR)in 70 cases of NPC tissues and 30 cases of nasopharyngeal mucosa chronic inflammation tissues.The differential expression levels between NPC tissues and nasopharyngeal chronic inflammatory tissues were analyzed,and the correlation among their expression levels in NPC tissues and patients’ clinicopathological characteristicswere evaluated and Disease-free survival time.Results:(1)The expression of EBV-miR-Bart1-3p in NPC tissues was significantly higher than that in nasopharyngeal mucositis tissues(P<0.05);the expression level of EBV-miR-Bart1-3p in NPC tissues was not significantly associated with patients’ sex,age,clinical stage,lymph node metastasis at initial diagnosis and Disease-free survival time(P> 0.05).(2)There was no significant difference between hsa-miR-23b-3p expression and nasopharyngeal mucositis tissues(P>0.05).There was no significant correlation between the hsa-miR-23b-3p expression level in NPC tissues and patients’ sex,age,clinical stage,lymph node metastasis at initial diagnosis and Disease-free survival time(P>0.05).Conclusions:(1)The expression level of EBV-miR-Bart1-3p in NPC tissues is higher than that of chronic inflammatory tissues of the nasopharynx mucosa,and it is speculated that it may be involved in the occurrence of NPC.(2)hsa-miR-23b-3p may take little contributions on NPC carcinogenesis. |
