Objective: To determine the expression of miR-1973 and miR-2113 in nasopharyngeal carcinoma(NPC)tissues and nasopharyngeal mucosal chronic inflammation tissues,respectively,and to analyze the relationship between their expression levels and distant metastasis and clinical prognosis of NPC.Methods: The expression levels of miR-1973 and miR-2113 in NPC tissues and nasopharyngeal mucosal chronic inflammation tissues were detected by quantitative real-time PCR(q RT-PCR),respectively,and their expression intensity was analyzed in relation to the clinicopathological parameters of NPC;Kaplan-Meier survival curve study was used to analyse the effect of miR-1973 and miR-2113 expression on the 3-year overall survival of NPC patients;ROC curve was used to analyse the predictive value of miR-1973 and miR-2113 in the diagnosis of NPC.Results: The expression level of miR-1973 in NPC tissues was significantly higher than that in tissues with chronic inflammation of the nasopharyngeal mucosa,and the difference was statistically significant(P < 0.05);the intensity of miR-1973 expression was significantly correlated with distant metastasis of NPC,and the difference was statistically significant(P <0.05)).The overall survival rate at 3 years was significantly higher in the low miR-1973 expression group than in the high miR-1973 expression group,and the difference was statistically significant(P < 0.05);the receiver operating characteristic(ROC)curve showed that the area under the miR-1973 expression diagnosis The area under the curve(AUC)of the NPC curve was 0.9647.The expression level of miR-2113 was significantly upregulated in NPC tissues compared with that of chronic inflammation of the nasopharyngeal mucosa,and the difference was statistically significant(P < 0.05).miR-2113’s relative expression level did not correlate with patient age,gender,clinical stage,presence of lymph node metastasis at initial diagnosis,and presence of distant metastasis at initial diagnosis,and the difference was not statistically significant(P > 0.05).ROC curve showed that the AUC of miR-2113 expression for the diagnosis of NPC was 0.9283.Conclusion: The relative expressions of miR-1973 and miR-2113 were significantly upregulated in NPC tissues,presumably they may be involved in the development of NPC;miR-1973 was positively correlated with distant metastasis of NPC,presumably it may promote the progression of NPC;miR-1973 expression was closely related to prognosis and may be a molecular marker for predicting the prognosis of NPC;miR-1973 and miR-2113 have superior diagnostic efficacy for NPC. |
