| ObjectiveBased on the autism spectrum disorder "drug-target" association network constructed in previous studies,the relationship between autism-related drugs and targets was predicted.The study found that the drug amitriptyline may be effective in the treatment of autism spectrum disorder.In this study,bioinformatics methods were used to analyze the effect of amitriptyline on genome-level expression profiles,in order to explore whether amitriptyline has a potential therapeutic effect on autism spectrum disorders,so as to provide basic research on autism spectrum disorders.and clinical treatment.Data and MethodsThe genome-wide expression profiling data and control data of amitriptyline acting on rat kidney were obtained from the Gene expression omnibus(GEO),and R,Venny and String were used to perform differential analysis,GO enrichment analysis,KEGG enrichment analysis.Set analysis,protein interaction network analysis(PPI)and key gene screening;then use "Autism" as the key word to search related data in the GEO database,and conduct differential analysis and pathway enrichment analysis to prove the effect of amitriptyline There is a correlation between the pathway and the action pathway of autism spectrum disorder.The domain knowledge scoring method was used to verify the effectiveness of amitriptyline-related targets,and the "drug-disease" association analysis was performed using the apparent precision treatment prediction platform to calculate negatively related diseases,positively related drugs and related in vivo targets.ResultA total of 331 amitriptyline differential genes were screened,of which 179 were up-regulated and 152 were down-regulated.A total of 393 differential genes were screened in the autism spectrum disorder dataset,of which 165 were up-regulated and 228 were down-regulated.The enrichment analysis found that GO enrichment reactions were mainly involved in protein phosphorylation,inflammatory response and immune response.The enrichment of KEGG pathway is mainly related to oxidative phosphorylation reaction,morphine or cocaine addiction,MAPK signaling pathway,Huntington’s disease and Alzheimer’s disease and other mechanisms.Through text verification,a total of 10 literatures related to autism spectrum disorder and the role of key targets were retrieved,verifying the validity of relevant key targets.Combined with the computational results of Epimed,a clinical bioinformatics apparent precision treatment prediction platform,it was found that amitriptyline may have potential therapeutic effects,including autism spectrum disorder,idiopathic pulmonary fibrosis,type 2 diabetes,Huntington’s chorea disease,neurofibromatosis,etc.It is predicted that amitriptyline combined with western medicines resveratrol,phenisotoin,naltrexone,and traditional Chinese medicines such as cordyceps sinensis,acacia,uncaria,musk,etc.have potential synergistic effects on mental health diseases,which are worthy of further in-depth research.ConclusionBased on genome-wide expression profiling data,using differentially expressed gene fold method,cluster analysis method,protein interaction network analysis method,etc.,and with the help of bioinformatics analysis,it was found that amitriptyline has a potential therapeutic effect on autism spectrum disorder. |
