Objective As a natural green medicinal plant with rich wild resources,Viburnum sargentii can treat a variety of diseases and has broad application prospects.The purpose of this study was to systematically analyze the chemical components of the medicinal parts of Viburnum sargentii by using Ultra-High Performance Liquid Chromatography High-resolution Mass Spectrometry(UHPLC-MS)as the test platform,and to analyze the differences of the chemical components of the branches,leaves and fruits of Viburnum sargentii.Methods The extraction conditions were optimized by single factor and star point design,and the phytochemical components in the Viburnum sargentii were extracted by condensation reflux extraction method.The chemical components of the aqueous extracts from the tender branches,leaves and fruits of the three medicinal parts of Viburnum sargentii were identified by UHPLC-MS combined with the database and compound discoverer 3.2 software.At the same time,the contents of three active components were determined by liquid chromatography-mass spectrometry.Finally,SPSS software was used to analyze the principal components of the medicinal parts of Viburnum sargentii to judge the differences of chemical components in its twigs,leaves and fruits.Results Taking the number of extracted compounds as the goal,through single factor and star point design,the optimal extraction conditions for extracting the tender branches of Viburnum sargentii were optimized.The optimal extraction conditions were condensation reflux extraction method,the solid-liquid ratio was 1:154.5 and the condensation time was 125.5 min.A total of 170chemical components were identified from the water extract of Viburnum sargentii(twigs,leaves and fruits).Compounds with a matching degree of more than 80%were identified by compound discoverer 3.2 software.A total of 85 active components were identified,including 12 amino acids,16 flavonoids and glycosides,4 phenylpropanoids,3 alkaloids,6 phenolic acids,2 coumarins and 2esters,22 organic acids and 18 other compounds.Among the 85 active ingredients,18 unique compounds were identified from twigs,18 unique compounds were identified from leaves,21unique compounds were identified from fruits,and 28 compounds were identified from the three medicinal parts.In this plant,the content of chlorogenic acid is relatively high,and the content of amino acids is also high.There are obvious differences in the types and contents of chemical components in different parts.The content determination results showed that the contents of Chlorogenic acid,Catechin and L-phenylalanine in the tender branches of Viburnum sargentii were0.182 mg·g-1,0.172 mg·g-1and 0.215 mg·g-1respectively;The contents in leaves were 0.351mg·g-1,0.273 mg·g-1and 0.256 mg·g-1respectively;The contents in fruit were 0.507 mg·g-1,0.305 mg·g-1and 0.107 mg·g-1respectively.The results of difference analysis showed that there were 8 kinds of multi amino acids and 6 kinds of unique amino acids in twigs.There are 10 kinds of organic acids in leaves and 4 kinds of unique organic acids.There are more flavonoids in fruits,including 10 kinds and 9 kinds of unique flavonoids.There are also obvious differences in the content of the same components.Conclusions In this paper,a comprehensive qualitative and quantitative study was carried out on the chemical components in the water extract of twigs,leaves and fruits.The results show that there are many kinds of chemical components in the Viburnum sargentii,and the contents of active components Chlorogenic acid,Catechin and L-phenylalanine are high.The Viburnum sargentii is extremely rich in resources in China,which is a good greening and ornamental plant,At the same time,it also contains a large number of bioactive components,so it has high development value.It is helpful to understand the chemical basis of the plant and realizing its economic value in the field of medicine and health care.
