| Background and Objective:Centromere protein U(CENPU),as one of the critical resident centromere-associated proteins,is an essential component of the inner layer of the kinetochore to form the CCAN network(constitutively centromere-associated network,CCAN).One,involved in the assembly of the mature kinetochore in metaphase,plays an essential role in cell mitosis.Recent studies have found that CENPU expression is closely related to malignant tumors.This study intends to clarify the role and mechanism of CENPU in the proliferation,apoptosis,and migration of gastric cancer cells through in vivo and in vitro experiments and clinical case data research,enrich the current understanding of the function of the CENPU gene and provide a basis for the research on the occurrence and development of gastric cancer,Provide new targets.Method:1.Using bioinformatics combined with gastric cancer data set in TCGA database and normal gastric tissue data set in GTEX database CENPU expression level and related patient prognosis,using immunohistochemistry,RT-PCR and other techniques to detect CENPU protein in gastric cancer.Expression levels in tissues.2.After down-regulating the expression of CENPU in AGS,Ed U experiments showed that its proliferation ability was weakened;flow cytometry results showed that AGS cells were blocked in the G1 phase,and the proportion of apoptosis increased;migration-related experiments showed that their migration ability was weakened;Tumor experiments also confirmed that the tumor body was significantly reduced after stably down-regulating the expression level of CENPU.3.Compared with sh NC/AGS,the expression levels of PI3 K,AKT,and PTEN proteins in sh CENPU/AGS decreased,the expression levels of proliferation-related proteins PCNA proteins decreased,and the expression levels of apoptosis-related proteins BAX and BCl-2 proteins decreased.Result:1.The high expression of CENPU in gastric cancer tissue was analyzed by bioinformatics and was associated with poor prognosis of patients.The detection of IHC,RT-PCR,and other techniques showed that the expression of CENPU in clinical specimens of cancer tissue was higher than that in adjacent tissue(p< 0.05).2.After down-regulating the expression of CENPU in AGS,Ed U experiments showed that its proliferation ability was weakened;flow cytometry results showed that AGS cells were blocked in G1 phase,and the proportion of apoptosis increased;migration-related experiments showed that their migration ability was weakened;Tumor experiments also confirmed that the tumor body was significantly reduced after stably down-regulating the expression level of CENPU.3.Compared with sh NC/AGS,the expression levels of PI3 K,AKT,and PTEN proteins in sh CENPU/AGS decreased,the expression levels of proliferation-related proteins PCNA proteins decreased,and the protein expressions of apoptosis-related proteins BAX and BCl-2 decreased.Conclusion:1.Through bioinformatics analysis and clinical sample verification,it is shown that the expression of CENPU in gastric cancer is related to the poor prognosis of patients,and it may become an indicator for predicting the prognosis of gastric cancer patients.2.CENPU is a cancer-promoting factor in gastric cancer.In vivo and in vitro experiments have confirmed that CENPU affects the proliferation of gastric cancer cells.At the same time,in vitro experiments demonstrated that CENPU can regulate gastric cancer cells’ apoptosis and migration ability.3.CENPU regulates the proliferation,apoptosis,and migration of gastric cancer cells through the PI3K/AKT pathway. |
