Manganese is a necessary trace element for the normal development of the human body.However,excessive exposure to manganese can lead to innate immune inflammatory response and cause inflammatory damage.Understanding the mechanism of human immune response to manganese is helpful to explore the ways to control the toxic damage in the early stage.As a protein complex,inflammsome regulate the immune response by releasing inflammatory cytokines.NLRP3 inflammsome play a key role in a variety of physiological processes related to the immune system.At present,the studies on the toxicity of manganese at home and abroad are mostly focused on neuroinflammatory damage,but there are few studies on peripheral immune damage of manganese,and there is a lack of research on the correlation of organ and tissue damage caused by manganese exposure in mice.Therefore,this study takes Mn Cl2as an example to explore the mechanism of innate immune damage caused by manganese,so as to provide effective measures for further prevention and treatment.The purpose of this study was to explore the role and mechanism of NLRP3 inflammsome in the innate immune response of peritoneal macrophages in C57BL/6 mice induced by manganese exposure and the relationship between NLRP3 inflammsome and organ and tissue damage induced by manganese exposure.The results show that peritoneal macrophages exposed to manganese can be activated and induce immune inflammation,which is mainly mediated by NLRP3 inflammsome.At the same time,the results of in vivo experiments showed that chronic manganese exposure could cause damage to the nervous system and organs of mice.To sum up,manganese exposure will cause macrophages to release inflammatory factors,damage the immune function of the body,induce inflammatory response,and further damage tissues and organs...
