| Objectives:By studying the expression levels of FTO and IGF2BP2 gene transcription products(mRNA),serum protein and the changes of clinical physicochemical indexes in patients of the liver,spleen,and kidney with the common disease locations of phlegm syndrome of Metabolic Syndrome(MS),the genetic mechanisms and possible material bases of common disease locations of phlegm syndrome of MS from the perspectives of molecular biology were preliminary elucidated,the scientific connotations of the pathogenesis theories of common disease locations of phlegm syndrome of MS were enriched,objective bases of the syndrome differentiation and treatments of MS were provided,and reference bases for the early diagnoses,clinical interventions and prognosis judgments of them were also provided.Methods : 1.The patients of MS who are conformed to the diagnostic standards and healthy people during this period were collected,basic data and four-diagnosis information of them were collected.The method of “syndrome elements differentiation” was used,thirty patients of liver,spleen and kidney with common disease locations of phlegm syndrome of MS were separately screened and thirty healthy subjects were used as controls,blood samples were collected,biochemical indexes were detected and serums were separated.2.Q-PCR and ELISA were used to detect the relative levels of FTO and IGF2BP2 genes mRNA and protein expression in every group,the differences of gene transcription levels and protein levels were analyzed.3.The correlations between FTO,IGF2BP2 genes and common disease locations of phlegm syndrome of MS and its physicochemical indexes were studied by Chi-square test,One-way ANOVA,Non-parametric test,Pearson correlation analysis and Spearman correlation analysis.Results : 1.Basic data in healthy group and MS phlegm syndrome liver,spleen and kidney groups(1)There were no statistical significances in gender composition among all groups(P>0.05).(2)Compared with the healthy group,the ages of MS phlegm syndrome liver and kidney groups were increased,the differences were statistically significant(P<0.01).2.Comparisons of clinical physicochemical indexes levels in healthy group and MS phlegm syndrome liver,spleen and kidney groups(1)BMI,DBP and TG: Compared with the healthy group,MS phlegm syndrome spleen and kidney groups increased in BMI,DBP and TG,the differences were statistically significant(P<0.05,P<0.01);compared with the MS phlegm syndrome liver group,MS phlegm syndrome spleen group increased in BMI,DBP and TG,MS phlegm syndrome kidney group increased in DBP,the differences were statistically significant(P<0.05,P<0.01).(2)WC,SBP and FPG: Compared with the healthy group,MS phlegm syndrome spleen and kidney groups increased in WC,SBP and FPG,MS phlegm syndrome liver group increased in WC and FPG,the differences were statistically significant(P<0.01).(3)HDL-C: Compared with the healthy group,MS phlegm syndrome spleen and kidney groups decreased,the differences were statistically significant(P<0.05,P<0.01);compared with the MS phlegm syndrome liver group,MS phlegm syndrome spleen group decreased,the difference was statistically significant(P<0.01).3.FTO and IGF2BP2 genes relative expression levels in healthy group and MS phlegm syndrome liver,spleen and kidney groupsCompared with the healthy group,the relative expression levels of FTO mRNA and IGF2BP2 mRNA in MS phlegm syndrome liver group were increased,the differences were statistically significant(P<0.01);compared with the MS phlegm syndrome liver group,the relative expression levels of FTO mRNA and IGF2BP2 mRNA in MS phlegm syndrome kidney group were decreased,the relative expression level of IGF2BP2 mRNA in MS phlegm syndrome spleen group was decreased,the differences were statistically significant(P<0.05,P<0.01).Compared with the healthy group,the relative expression levels of FTO protein in MS phlegm syndrome liver,spleen and kidney groups were decreased,the relative expression levels of IGF2BP2 protein in MS phlegm syndrome liver,spleen and kidney groups were increased,the differences were statistically significant(P<0.05,P<0.01).4.Correlations of the scores of phlegm syndrome element and the scores of disease locations syndrome elements,clinical physicochemical indexes,FTO gene and IGF2BP2 gene in common disease locations of MS phlegm syndrome(1)The score of phlegm syndrome element in healthy group was positively correlated with the score of liver syndrome element(P<0.01);the score of phlegm syndrome element in MS phlegm syndrome liver group was positively correlated with the score of kidney syndrome element(P<0.05).(2)The score of phlegm syndrome element in MS phlegm syndrome spleen group was positively correlated with WC and the relative expression level of IGF2BP2 mRNA(P<0.05,P<0.01);the score of phlegm syndrome element in MS phlegm syndrome kidney group was positively correlated with the relative expression level of FTO mRNA(P<0.05).5.Correlations in relative expression levels of FTO and IGF2BP2 genes and clinical physicochemical indexes of common disease locations of MS phlegm syndrome(1)The relative expression level of FTO mRNA in MS phlegm syndrome liver group was positively correlated with HDL-C,and negatively with FPG(P<0.05);the relative expression level of FTO mRNA in MS phlegm syndrome kidney group was negatively correlated with WC(P<0.05).(2)The relative expression level of IGF2BP2 mRNA in healthy group was positively correlated with WC(P<0.05);the relative expression level of IGF2BP2 mRNA in MS phlegm syndrome spleen group was negatively correlated with DBP(P<0.05).(3)The relative expression level of FTO protein in MS phlegm syndrome kidney group was positively correlated with LDL-C(P<0.05).(4)The relative expression level of IGF2BP2 protein in MS phlegm syndrome liver group was negatively correlated with DBP(P<0.01);the relative expression level of IGF2BP2 protein in MS phlegm syndrome kidney group was negatively correlated with WC,and positively correlated with SBP and TC(P<0.05).Conclusions:1.The formation of the common disease locations of MS phlegm syndrome had different biological bases.Among them,the formation of MS phlegm syndrome liver disease location may be related to the increase of relative expression levels of FTO and IGF2BP2 genes mRNA;the formation of MS phlegm syndrome spleen disease location may be related to BMI,TG,DBP and HDL-C;the formation of MS phlegm syndrome kidney disease location may be related to DBP.2.There were some correlations among syndrome elements,clinical physicochemical indexes,FTO and IGF2BP2 genes in common disease locations of MS phlegm syndrome.The formation of phlegm syndrome element in MS phlegm syndrome spleen disease location may be positively related to WC and the relative expression level of IGF2BP2 mRNA.The formation of phlegm syndrome element in MS phlegm syndrome kidney disease location may be positively related to the relative expression level of FTO mRNA.The relative expression level of FTO protein in MS phlegm syndrome kidney disease location may be positively related to LDL-C.The relative expression level of IGF2BP2 protein in MS phlegm syndrome kidney disease location may be positively related to SBP and TC. |
