| Objective: To compare the biological classification characteristics of intestinal flora between atopic dermatitis and healthy infants at age 6-12 months in Suining area based on the 16 S r DNA sequencing technique,so as to provide basis for the prevention and treatment of infantile atopic dermatitis.Methods: 1.Clinical investigation: 34 patients with atopic dermatitis who met the diagnostic criteria of Williams and 38 healthy controls excluding related allergic diseases were included.Fecal samples were collected at age 6-12 months.2.Detection and analysis of intestinal flora: a total of 72 qualified fecal samples taken from 34 atopic dermatitis and 38 healthy control children were collected,who were served as AD and HC group respectively.All fecal samples collected were sent to Tianjin High-throughput Sequencing Lab for the detection of bacteria,and the data were combined with clinical data.The characteristics of intestinal flora in patients with atopic dermatitis and healthy controls at age 6-12 months in Suining area were analyzed.Results: 1.Baseline of subjects: 34 patients with atopic dermatitis and 38 healthy babies were permanent residents of Suining.There was no significant difference in gender and age between the two groups(P>0.05).2.Detection and analysis of intestinal flora: a total of 72 qualified fecal samples were collected,which were divided into atopic dermatitis group(n=34)and healthy control group(n=38).The results of comparison between the two groups showed that:(1)Firmicutes,Bacteroidetes and Proteobacteria,Actinobacteria were the main bacteria,accounting for more than 95% of the total.The relative abundance of AD group was 37.58%,14.99%,19.95% and 22.97% respectively,and that of healthy group was 29.80%,14.13%,28.76% and 23.64%.The relative abundance of Firmicutes and Bacteroidetes in the AD group was higher than that in the healthy group,while that of Proteobacteria and Actinobacteria was lower than that in the healthy group,but there was no significant difference between the two groups(P>0.05).(2)Compare the microflora with the top ten at each level.At the Phylum level,the relative abundance of Aciclobacteriota in the AD group was significantly higher than that in the healthy control group(P<0.05).At the Class level,the relative abundance of Acidobacteriae and Alphaproteobacteria in the AD group was higher than that in the healthy group(P<0.05).At the order level,the relative abundance of Enterobacterales in the AD group was lower than that in the healthy group(P<0.05).At the family level,the relative abundance of Enterobacteriacea in the AD group was significantly lower than that in the healthy group(P<0.05).And at the genus level,the relative abundance of Escherichia-Shigella in the AD group was significantly lower than that in the healthy group(P<0.05),and at the species level,the relative abundance of Escherichia-coli in the AD group was significantly lower than that in the healthy group(P<0.05).(3)there was no significant difference between patients with atopic dermatitis and healthy babies by comparing the important probiotics such as Bifidobacterium,Lactohacillus and Akkermansia.Conclusion: There is no significant change in the main intestinal flora in infants with atopic dermatitis at age 6-12 months in Suining area,but Enterobacterales,Enterobacter-iaceae and Escherichia-Shigella,Escherichia-coli decrease significantly,which may be related to atopic dermatitis. |
