Objective: To investigate the correlation between IL-22,neutrophil and the severity of chronic sinusitis nasal polyp(CRSw NP)by detecting the expression level of IL-22 and the degree of neutrophil infiltration,so as to provide new experimental data for the diagnosis and treatment of chronic sinusitis(CRS).Methods: From November 2019 to September 2020,36 patients hospitalized in the department of Otolaryngology,head and neck Surgery of a public grade A hospital and diagnosed with CRSw NP before surgery were recruited as the CRSw NP group(experimental group),and 12 patients admitted for surgical treatment with nasal septum deviation were recruited as the control group at the same time.General information and blood routine reports of all enrolled patients were collected before surgery,and relevant data were counted.The Sino-Nasal Outcome test-22(SNOT-22)scale,the Lund-Mackay scale,and the Lund-Kennedy scale were used to score the patients,and venous blood was collected from each group at 7:00 a.m.the next day after admission.During the operation,the nasal polyp tissue of the CRSw NP group and the mucosal tissue of the inferior turbinate of the control group were collected,and divided them into two parts.One part used Hematoxylin-eosin staining(HE)to conduct inflammatory cell counting.The other part was used Enzyme Linked immunosorbent assay(ELISA)to detect IL-22 in serum and tissues of both groups.Patients were followed up 1 year after surgery,and the SNOT-22 scale and the Lund-Kennedy scale were used for postoperative evaluation.After collecting all experimental data,statistical software was used for statistical analysis.Results: 1.Compared with the control group,preoperative scales’ scores,percentage of tissue neutrophils and IL-22 concentration,postoperative SNOT-22 and Lund-Kennedy scales’ scores in the CRSw NP group were significantly higher(P<0.05).2.The rate of neutrophil in peripheral blood of the CRSw NP group was much higher than that of the control group(P<0.05),but there was no correlation with preoperative and postoperative scales’ scores of patients(P>0.05),and there was no difference in serum IL-22 concentration between the CRSw NP group and the control group(P>0.05).3.The percentage of neutrophil in the CRSw NP group was positively correlated with preoperative scales’ scores(P<0.05).The correlation was more significant in the non-eosinophilicchronic rhinosinusitis with nasal polyps(Non-e CRSw NP)group.4.Compared with the control group,IL-22 concentration in the CRSw NP group was positively correlated with preoperative scales’ scores(P<0.05),the correlation was more significant in the Non-e CRSw NP group.And with a weak positive correlation with the percentage of neutrophils(P<0.05).5.One year after surgery,IL-22 concentration and neutrophil percentage in the Non-e CRSw NP group were positively correlated with SNOT-22 and Lund-Kennedy scales’ scores(P<0.05).Conclusions: 1.The degree of neutrophil infiltration and IL-22 expression level in the CRSw NP group were both related to the severity of the disease and the postoperative improvement of the condition,and the correlation was stronger in the Non-e CRSw NP group.2.There was no major difference in serum IL-22 concentration between the two groups(P>0.05),suggesting that IL-22 may only play an inflammatory role in the local sinus,but does not cause systemic inflammation.
