Explore The Medication Law Of Doctors In Ming And Qing Dynasties And Professor Minghe Zhang In The Treatment Of Psoriatic Arthritis Based On Network Pharmacology

Study one:Data mining analysis of literature for treating psoriatic arthritis by physicians in Ming and Qing Dynasties.Objective:by collecting and analysing datas mining of ancient literatures of physicians in Ming and Qing Dynasties for the treaments of psoriatic arthritis,summarize the rules of medicine selected by physicians during the Ming and Qing Dynasties.Methods:Searching the literature consistent with symptoms of psoriatic arthritis included in the “Chinese Medical Dictionary”.Based on slection and exclusion criterion,and uniform standardization of data,and Establish a prescription database.The medical record datebase of the medical record cloud platform uses the function of data analysis to analysis compatibility law of traditional Chinese Medicine,and to provide a syndrome differentiation and treatment direction for current contemporary physicians treating psoriatic arthritis.Results:Through searching 23 medical records for the treament of psoriatic arthritis are included.High frequency use of traditional Chinese Medicine include Fangfeng,Gancao,Baizhu,Xixin,Qianghuo and Tianma.From the point of view of the nature of the medcine and the neridian of nature and taste,pungen、warm and bitter medicines are commonly used,which are classfied as the liver,lungs and spleen meridian.The core paired-compoents are Gancao and Fangfeng,Xixin and Fangfeng,Baizhu and gancao,Xixin and Gancao,Qianghuo and Fangfeng,Danggui and Fangfeng,Chuanxiong and Gancao,Tianma and Fangfeng.The result of the cluster analysis shows that the prescription can be divided into four groups,the first group is Gancao and Baizhu,the second group is Jingjie,Danggui and Qinjiao,the third group is Qianghuo and Tianma,the fourth group is Xixin and Chuanxiong.Conclusion: Through the analysis of ancient medical prescriptions,the physicians during the Ming and Qing Dynasties believed that the occurance of psoriatic arthritis is related to the intergrated result of the six exogenous factors.In terms of the rule of the law,the physicians during the Ming and Qing Dynasties follow therapeutic principle of warming and activating blood circulation,and choose drugs for relieving dampness,invigorating spleen and Supplementing Qi,which provides new ideas for modern and contemporary doctors to treat psoriatic arthritisStudy Two:Research on the mechanism of core paired-compoents in the treatment of psoriatic arthritis by doctors in the Ming and Qing Dynasties and Professor Minghe Zhang based on network pharmacologyObjective:Study on the molecular mechanism of core paired-compoents for the treatment of psoriatic arthritis by doctors in the Ming and Qing Dynasties and Professor Minghe Zhang based on network pharmacology.Methods:The active components and Effective target of Danpi,Lianqiao,Jinyinhua,Tufuling,Baizhu,Gancao,Fangfeng and Xixin are collected and screened through the TCMSP.The effective target was imported into Uni Prot database to standardize the name of target gene.The disease targets of Ps A are obtained from Drug Bank、Gene Cards and OMIM databases.Structuring the Venny diagram through Venny.2.1 website.The “traditional Chinese medicine-Drug active ingredient-target”and protein interaction of key proteins are obtained according to Cytoscape3.8.2 software,which is used for visualization research and network topology analysis.DAVID is used to carry out GO and KEGG analysis for the key targets,and the bubble and bar graphs of enrichment are made by R language.Results:1.A total of 1241 targets of psoriatic arthritis were obtained.Through the analysis of Professor Minghe Zhang core paired-compoents for the treatment of psoriatic arthritis,“ Jinyinhua-Lianqiao ”paired-compoents contains 46 active ingredient and 744 targets.KEGG pathway mainly includes HIF-1,NF kappa B and PPAR signaling pathway.“Jinyinhua-Tufuling”paired-compoents contains 38 active ingredient and 598 targets.KEGG pathway mainly includes NF kappa B and Wnt signaling pathway.“ Danpi-Lianqiao ” paired-compoents contains 34 active ingredient and584 targets.KEGG pathway mainly includes TNF、NF kappa B and Wnt signaling pathway.In MF items,there are enzyme blinding、indentical protein blinding and protein blinding.etc.In Bp items,there are positive regulation of transcription from RNA polymerase Ⅱ promoter、positive regulation of gene expression and response of drug.etc.In CC items,there are extracellular space、extracellular region and cytosol.ect.2.Through the analysis of physicians of Ming and Qing Dynasties core paired-compoents for the treatment of psoriatic arthritis,“Fangfeng-Gancao”paired-compoents contains 111 active ingredient and 197 targets.KEGG pathway mainly includes NF kappa B、IL-17 and JAK-STAT signaling pathway.“Xixin-Fangfeng”paired-compoents contains 26 active ingredient and 62 targets.KEGG pathway mainly includes NF kappa B、HIF-1 and AMPK signaling pathway.“Baizhu-Gancao”paired-compoents contains 93 active ingredient and 90 targets.KEGG pathway mainly includes NF kappa B、MAPK、PPAR and JAK-STAT signaling pathway.In MF items,there are DNA-binding transcription factor binding、kinase binding and protein homodimerization activity.etc.In Bp items,there are response to inorganic substance、hormone and positive regulation of cell migration.etc.In CC items,there are transcription regulator complex、membrane raft and vesicle lumen.ect.Conclusion:Minghe Zhang’s core paired-compoents for the treatment of psoriatic arthritis,“ Jinyinhua-Lianqiao ”、“ Jinyinhua-Tufuling ” and“Danpi-Lianqiao”,which could Inhibit inflammatory pathway and improve hormone level to alleviate joint inflammation and strengthen the osteogenesis function interval,so as to play a role in the treatment of psoriatic arthritis.Physicians of Ming and Qing Dynasties core paired-compoents for the treatment of psoriatic arthritis,“ Fangfeng-Gancao ”、“ Xixin-Fangfeng ” and“Baizhu-Gancao”,which could block inflammatory cytokines to inhibit skin keratinocyte metaplasia and alleviate skin lesions.Based on the system biology discipline of network pharmacology,it emphasizes the multi-channel regulation of traditional Chinese medicine on the multi signal pathway of disease,which provides the research basis of molecular mechanism for the further study of drugs for the treatment of PSA.
