Study On Network Pharmacology Mechanisms Of Jiawei Chaihu Shugan Decoction In The Treatment Of Epilepsy And Depression Based On The Concept Of "Treating Different Diseases With Same Method"

Objective: This study explored the effective active components,potential targets and related pathways of various traditional Chinese medicines of the modified Chaihu Shugan decoction with the help of network pharmacological methods,Then explain the potential mechanism of epilepsy and depression of "treating different diseases together" of Jiawei Chaihu Shugan decoction,so as to provide reference basis for follow-up in-depth research,clinical prescription selection and application.Methods: By searching the pharmacology database and analysis platform of traditional Chinese medicine system(TCMSP),taking the oral availability(OB)≥ 30% and drug like(DL)≥ 0.18 as the screening conditions,the compound components and possible therapeutic targets of the formula of Jiawei Chaihu Shugan decoction were obtained,and the Uni Prot database was used to predict the potential action targets of the active components.Secondly,we collected disease targets related to epilepsy and depression in combination with Gene Cards、Drug Bank、OMIM 和 Pharm Gkb databases,With the help of R software,integrate the above four databases to obtain the disease-related targets of the two respectively,and take the intersection between the drug target and the two disease targets to obtain the intersection target genes of the three.And use Cytoscape 3 9.1 software construction of drug active ingredient target disease relationship network diagram.The intersection target genes were introduced into the string database to obtain the relationship of protein-protein interaction and construct PPI subnetwork.Thirdly,go function enrichment analysis and KEGG pathway enrichment analysis of intersection target genes were carried out with the help of relevant packages in R language software.Results: through the network pharmacological analysis,154 compounds and244 effective active components corresponding to the target of Jiawei Chaihu Shugan decoction were screened;2182 and 3140 targets of epilepsy and depression were obtained in gene cards,drug bank,OMIM and pharm GKB databases respectively;79 intersection targets of diseases and drugs were drawn by Wayne diagram;Through Cytoscape 3 9.1 the platform constructs PPI sub network.CAT、TP53、CASP3、AKT1、MAPK1、TIMP1、MMP2、SERPINE1、CCL2、NR3C1、MAPK3、IL1B、STAT3、APP、FOS、CXCL8、GSK3B、PTGS2、MMP9、AR、IGFBP3、CREB1、ESR1、HIF1A、MYC and other 25 targets are obtained and determined as the main core targets.The results of go function enrichment analysis showed that the targets of Jiawei Chaihu Shugan decoction in the intervention of epilepsy and depression were mainly concentrated in 1975 biological processes,such as response to radiation,response to metal ions,multicellular biological process,response to light stimulation and response to lipopolysaccharide.Including membrane raft,membrane microdomain,postsynaptic membrane,plasma membrane raft and endoplasmic reticulum lumen,72 play an important role in cell components.In addition,144 such as nuclear receptor activity,ligand activated transcription factor activity,DNA binding transcription factor binding,RNA polymerase II specific DNA binding transcription factor binding,protease binding and phosphatase binding also account for a large proportion in terms of molecular function.A total of 173 metabolic pathways were obtained by KEGG pathway enrichment analysis.Among them,lipid and atherosclerosis,Posey’s sarcoma associated herpesvirus infection,AGE-RAGE signaling pathway in diabetes complications,hepatitis B,human cytomegalovirus infection,IL-17 signaling pathway,prostate cancer,colorectal cancer,endocrine resistance are the major signaling pathways regulating epilepsy and depression,and related to inflammatory response,apoptosis,immune response,oxidative stress and other processes.Conclusion: Under the guidance of the theory of "Treating Different Diseases with Same Method " in Traditional Chinese medicine,the results of this study show that naringin,quercetin,isorhamnetin β Sitosterol,paeoniflorin,kaempferol,stigmasterol and luteolin may be the main effective compounds of modified Chaihu Shugan decoction.It is speculated that these active ingredients act on the reaction process of metal ions,oxidative stress response,through regulating TP53,CASP3,AKT1,MAPK1,MAPK3,IL1 B,FOS,CCL2,STAT3,MMP9,and MMP2 core hub nodes Many biological processes such as regulating lipopolysaccharide,protease binding and phosphatase binding affect the function of AGE-RAGE signaling pathway such as lipid and atherosclerosis,diabetes complications,human cytomegalovirus infection,IL-17 signaling pathway,cancer pathway and other common signaling pathways,resulting in mediating inflammatory cytokines and neurotrophic factors,and regulating synaptic excitability of neurons.It is revealed that Jiawei Chaihu Shugan decoction regulates the occurrence and development of epilepsy and depression through multi-component,multi-target and multi-pathway mechanisms,and plays the therapeutic effect of treating different diseases with the same way.
