| Objective: through the method of network pharmacology,to screen the main effective components of Yiqi Huoxue Kaixin Decoction in the treatment of coronary heart disease combined with depression,speculate its effective target,construct the network of traditional Chinese Medicine-component-target-disease,and explore the mechanism of Yiqi Huoxue Kaixin Decoction in the treatment of coronary heart disease combined with depression,so as to provide a theoretical basis for further experimental research.Methods: the compounds and effective targets of Yiqi Huoxue Kaixin Decoction were collected by tcmsp traditional Chinese Medicine System Pharmacology database,and the compounds and effective targets of Polygala tenuifolia were collected by ETCM traditional Chinese medicine website;Using genecards and OMIM database to screen disease targets,take the intersection and draw Venn diagram;Cytoscape software was used to construct the drug active ingredient key target network;The target protein interaction(PPI)network was constructed from sting database;The dose,clusterprofiler and pathview r packages were used to analyze the enrichment of target go function and KEGG pathway,predict its action mechanism,and draw histogram and Bubble Diagram to visualize the results.Results:(1)Active ingredients of traditional Chinese medicine: 205 active compounds,267 drug targets,1612 disease targets and 137 intersection targets were obtained.(2)Network key target prediction may include AKT1,IL6,TNF,VEGFA,IL1 B,CASP3,TP53,EGFR,MMP9,etc.(3)Go function analysis the mechanism of Yiqi Huoxue Kaixin Decoction in the treatment of coronary heart disease complicated with depression.There are 2705 go items,2405 biological process items,109 cell composition items and 191 molecular function items.(4)Through the enrichment analysis of KEGG pathway,183pathways were obtained,including Akt signaling pathway and atherosclerosis signaling pathway,VEGF signaling pathway,IL-6 signaling pathway,TNF signaling pathway,etc.Conclusion: Quercetin,luteolin,kaempferol,tanshinone,formononetin and other core components act on AKT1,IL6,TNF,VEGFA,IL1 B and other core genes,play biological roles such as inflammatory response,apoptosis,cell proliferation and oxidative stress,and finally regulate Akt signaling pathway and atherosclerosis signaling pathway,VEGF signaling pathway,IL-6signaling pathway TNF and other pathways play a role in the treatment of CHD complicated with depression. |
