| In a rapidly developing society,fatigue has become a common social problem,affecting people’s work efficiency and quality of life.Fatigue refers to the inability of the body to maintain a predetermined exercise intensity at a certain level,and the most direct performance is the decline of exercise endurance.Fatigue can be divided into exercise fatigue(mainly peripheral tissue fatigue)and central fatigue(mainly central nervous system abnormalities).Vine tea is a different tea from the young stems and leaves of Ampelopsis grossedentata(Hand.-Mazz.)W.T.Wang,a vine plant of the Ampelopsis genus.It has a long drinking history in southern of China,which has good effects on hypoglycemic,hypolipidemic,antioxidant,anti-inflammatory and other.Vine tea is rich in dihydromyricetin(DMY,up to 40%dry weight),in recent years,studies have found that natural plant products,such as flavonoids,play an important role in improving the fatigue.DMY is a natural flavonoid with a wide range of pharmacological activities.At present,there are a few studies reporting that DMY can relieve fatigue,but the research on its anti-fatigue mechanisms is almost blank.so we speculate that vine tea may also have the effect of improving fatigue.The purpose of this paper is to study the anti-fatigue effect and mechanism of vine tea and DMY,and provide the basis for their further research,development and application.(1)The anti-fatigue effect of DMY was studied by the weight-loaded swimming experiment in mice.The results showed that DMY could significantly prolong the exhaustive time of weight-loaded swimming in mice,reduce the content of serum LD and SUN,reduce the activities of serum LDH and CK,increase the glycogen reserve,and improve the activity of liver GSH-Px.By the results of qRT-PCR and western blot showed that DMY may improve fatigue of mice by activating muscle AMPK pathway and liver Nrf2,PPARα related pathways.(2)The anti-fatigue effect of vine tea was studied by weight-loaded swimming experiment,and the results showed that vine tea water extract could significantly improve the fatigue of mice.The results of further molecular mechanism studies suggested that regulating muscle mass and reducing muscle damage through PI3K-Akt-FoxO may be the key pathway for vine tea to exert its anti-fatigue effect.(3)The 21-day chronic sleep deprivation-induced central fatigue mouse model was further used to study the effect of vine tea on improving the fatigue.The results of the open field test and elevated plus maze test showed that compared with the normal group.the model group had obvious anxiety-like emotional tendencies.The anxiety was significantly improved after the intervention of vine tea extract.The results of serum LD,SUN and LDH in mice indicated that vine tea improved peripheral energy metabolism and physical function in central-fatigue mice.Joint analysis of transcriptomics and metabolomics in the hippocampus of mice suggested that vine tea may relieve anxiety caused by central fatigue mainly by mediating the function of serotonergic neuron system and its downstream PI3K-Akt and MAPK signaling pathway,and the specific mechanism needs to be further explored.In conclusion,this paper is the first to discuss the effect and potential molecular mechanism of vine tea on improving exercise fatigue,and the differences of mechanisms between vine tea and dihydromyricetin deserves further study.In addition,this paper finds for the first time that vine tea can significantly relieve anxiety in mice with central fatigue.The research promotes the research and development of vine tea in ethnic medicines,and is of great significance for the high-value development and healthy utilization of vine tea. |
