Effect Of Xiaojianzhong Decoction On Skeletal Muscle Of Exercise Fatigue Mice Based On AMPK/PGC1-α Pathway

Objectives:1.To explore the effects of Xiaojianzhong Decoction on the energy and material depletion,accumulation of metabolites,and internal environment imbalance in mice by establishing a running exhaustion model for mice,and to clarify Xiaojianzhong Decoction’s anti-fatigue mechanism;2.To observe the effect of Xiaojianzhong Decoction on energy metabolism of exhausted mice by immunoblot test,and to explore its role in AMPK/PGC1-αsignaling pathway.Methods:Forty male Kunming mice were randomly divided into normal group,model group,Xiaojianzhong decoction low dose group(XJZTD group),Xiaojianzhong decoction medium dose group(XJZTZ group),Xiaojianzhong decoction high dose group(XJZTG group)And Sijunzitang group(SJZT group),10 in each group.The model group,Xiaojianzhong decoction(low,middle,high)group and Sijunzitang group were trained on a 6-day treadmill to establish a fatigue model.The model group was given the same amount of normal saline.Xiaojianzhong decoction(low,middle,high)Gavage was given(2.5,5,10 g·kg-1),and the Sijunzi Decoction group was given 1 g·kg-11 for 6 consecutive days.After the experiment,the weight of each group of mice and the time of exhaustion after running were measured,and the serum urea(UREA),creatine kinase(CK),and lactate dehydrogenase(LDH)of each group were measured by colorimetric method,Muscle glycogen(MG),Na+-K+-ATPase,Ca2+-Mg2+-ATPase activity in skeletal muscle;hematoxylin-eosin(HE)staining was used to observe the pathological changes of skeletal muscle in each group;Western-blot was used to determine the protein expression of AMPK and PGC1-αin each group of skeletal muscle.Results:1.Compared with the normal group,the weight of mice in the model group was significantly reduced(p<0.01);compared with the model group,the weight of the XJZTD,XJZTZ,and XJZTG groups was significantly increased(p<0.01),and the exhaustion time was significantly prolonged(p<0.01,p<0.05),the exhaustion time in the SJZT group was significantly prolonged(p<0.05).2.Compared with the normal group,the levels of UREA and CK in the model group were significantly increased(p<0.01),and the LDH content was significantly decreased(p<0.01);Compared with the model group,the levels of UREA and CK in the XJZTD,XJZTZ,and XJZTG groups were significantly higher.Decrease(p<0.01),LDH content increased significantly(p<0.01),UREA and CK content in SJZT group decreased significantly(p<0.01,p<0.05).3.Compared with the normal group,the content of Na+-K+-ATP,Ca2+-Mg2+-ATP,and MG in the model group decreased significantly(p<0.01,p<0.05);Compared with the model group,XJZTD,XJZTZ,and XJZTG groups The content of Na+-K+-ATP,Ca2+-Mg2+-ATP,and MG increased significantly(p<0.01),and the content of Ca2+-Mg2+-ATP increased significantly in the SJZT group(p<0.05).4.Compared with the normal group,the expression of AMPK and PGC1-αprotein in the model group increased significantly(p<0.01);Compared with the model group,the expression of AMPK and PGC1-αprotein in the XJZTD,XJZTZ,XJZTG and SJZT groups increased significantly(p<0.01).Conclusion:1.Xiaojianzhong decoction can increase the weight of mice,prolong the exhausted running time of fatigued mice,and enhance the glycogen reserves of mice;it can promote the synthesis of ATPase and improve the function of mitochondria,provide energy for skeletal muscle cells,and protect the integrity of muscle cell membranes Sex,prevent the production of lipid peroxides,improve the anti-fatigue ability of mice.2.Xiaojianzhong decoction can improve the anti-fatigue ability of mice,and its mechanism is related to the activation of AMPK/PGC1-αpathway to enhance the synthesis of energy metabolic enzymes,improve the clearance rate of metabolic products,and enhance the stability of the body’s energy substances.3.The Xiaojianzhong decoction group was better than the Sijunzi decoction group in multiple indicators of fatigue resistance,and the recovery state was close to the normal group in terms of promoting several indicators of metabolites and increasing glycogen reserves.It shows that the overwork injury causes the camp and guard to lose balance,the yin and yang disorder,and the Qi rebellion are closer to the nature of fatigue.At the same time,it also shows that Xiaojianzhong decoction reconciles the Yingwei,regulates yin and yang,and regulates the Qi Qi method,which is more advantageous in anti-fatigue and can quickly relieve fatigue than the simple Qi supplement method.
