To research the anti-obesity effect of panax notoginseng saponins(PNS),and preliminary study its mechanism of action.First,PNS(100,200,400 mg/kg)was given to normal mice for 14 days.The changes of body weight,24 hours food and water intake,fasting blood glucose,fed blood sugar and glucose tolerance were recorded.The changes of heat dissipation in the back of mice were detected by infrared thermography.RT-PCR was used to detect the expression of heat producing genes in various fat tissues of mice,including uncoupled protein 1(UCP1).In addition,the diets induced obesity(DIO)mice model induced by high fat diet(HFD)was used.PNS(200,400 mg/kg)was given for 9 weeks by gavage,and the weight of mice was recorded.The changes of food and water intake,fasting blood glucose,fed blood glucose,glucose tolerance and insulin tolerance were measured.The changes of heat dissipation in the back of mice were detected by infrared thermography.The fat distribution of mice was detected by nuclear magnetic resonance imaging and the organ coefficient was calculated at the end of the treatment.HE staining was used to observe the visceral fat tissue(VAT)The pathological changes of subcutaneous fat tissue(SAT)and brown fat tissue(BAT)were detected by immunohistochemistry.The results showed that PNs could reduce fasting blood glucose and non fasting blood glucose in normal mice,and increase the back heat dissipation of normal mice in dose dependence,and up regulate the expression of heat gene mRNA in VAT,sat and bat in normal mice,including Ucp1,Dio2,Adrb3,Cidea,Pgc-1α.HE staining showed that the volume of adipocytes decreased,and the expression of UCP1 in all adipose tissues was increased by immunohistochemistry.In addition,PNS significantly inhibited the weight growth of DIO mice,reduced fasting blood glucose and fed blood glucose,improved glucose tolerance and insulin sensitivity,and had no significant influence on food and water intake;significantly improved fat distribution,reduced fat accumulation of VAT and SAT,increased BAT content,reduced lipid-water ratio,and increased the back heat dissipation of DIO mice;In conclusion,PNS has the effect of anti-obesity and metabolic syndrome,and its mechanism is related to promoting brown fat production heat and white fat browning and increasing energy consumption.PNS,as a potential anti-obesity drug,has a good prospect of deep development and clinical application. |
