| Objective:In this study,lipopolysaccharide(LPS)was used as an inducer to stimulate microglia(BV2)to establish an in vitro inflammatory cell model of cerebral ischemia,and to explore the effects and mechanisms of Panax notoginseng saponins and their main chemical components Rg1,Rb1,R1,Rd and Re on secondary inflammatory response of ischemic stroke,so as to provide theoretical basis for clinical prevention and treatment of ischemic stroke.Methods:Lipopolysaccharide(LPS)was used as inducer to stimulate microglia(BV2)to establish inflammatory cell model of cerebral ischemia.MTS method was used to detect the growth curve of BV2 cells and the effect of drugs on cell survival.ELISA kit was used to detect the effects of Panax notoginseng saponins and its main chemical components Rg1,Rb1,R1,Rd,Re on the expression of inflammatory factors NO,TNF-a,IL-1beta.Western blot was used to detect the expression of IKBa,Atg5 and Beclin1 proteins.Real-time fluorescence quantitative PCR was used to detect the expression of NF-kappa Bp65,Notch1 and Hes1.The effects of Panax notoginseng saponins and their main components on the secondary inflammatory response of ischemic stroke were investigated in terms of NF-kappa B,Notch signaling pathway and autophagy.Results:1.Total saponins of Panax notoginseng and its main components can reduce the contents of NO,TNF-a and IL-1beta.2.Panax notoginseng saponins can inhibit the expression of NF-kappa B p65,IKBa protein Notch1 and Hes1 in the signal pathway of NF-kappa B and Notch.The main components of Panax notoginseng saponins have selectivity for their expression.3.Panax notoginseng saponins inhibit the expression of autophagic gene Beclin1 and Atg5,and the main components of Panax notoginseng saponins have selectivity for their expression.Conclusion:1.Panax notoginseng saponins and their main constituents have inhibitory effects on inflammation.2.The anti-inflammatory effect of Panax notoginseng saponins may be achieved by inhibiting NF-kappa BăNotch and cell autophagy,and the effects of different components are selective. |
