Objective The research aim to investigate the effect on epiphyseal development by astragaloside in rats and it attempt to confirm that astragaloside can promote differentiation and proliferation of epiphysis stem cells,keep the well biological characteristics of epiphysis stem cells and promote epiphyseal growth.Methods The experiment obtain epiphysis stem cells by the method of culture newborn SD rats in vitro,identify epiphysis stem cells by using immunohistochemical FGFR-3 dyeing,detect passage cell by MTT method testing and select the third generation cells as a follow-up study experimental intervention.Chosing astragaloside induce epiphysis stem cells in vitro culture,using Collagen type Ⅱ monoclonal antibody staining to test cell biological characteristics,using MTT to test the activity of Epiphysis stem cells.In addition,four weeks old healthy SD rats were used in this study.The animals were randomly divided into experimental group(A)and control group(B),24 ineach group(regardless of gender).The rats of the group A received astragalus injection in a dose of 8.0 g / kg once a day.The group B was injected with an equal volume of salin.Killing 12 rats after 3 and 5 weeks,it need to measure the length of tibia by vernier caliper,receive X-ray examination and compare to each other by the length of the tibia.Results The results proved that the cells in vitro cultured FGFR-3 staining positive,and the cell is epiphysis stem cells.It makes the speed of cell proliferation accelerate through microscope after induction with astragaloside.Optical density of experiment group higher than control group by MTT method testing.Collagen typeⅡmonoclonal antibody staining is positive.The length of the tibia specimen and the X-rays of experiment group longer than control group obviously in fifth week.Conclusion This research proved that epiphyseal stem cells which was inducted with astragaloside can secrete characteristic matrix normally and keep the well biological characteristics of epiphysis stem cells.In addition,astragaloside can promote epiphyseal growth and delay epiphyseal closure.
