Astragaloside IV(AS-IV)is one of the main chemical active components of Astragalus membranaceus,with extensive pharmacological activities.Astragaloside IV has protective effects on local focal cerebral ischemia and brain injury,cardiovascular disease,pulmonary disease,liver cirrhosis and diabetic nephropathy,and can improve organ fibrosis,inflammatory response,oxidative stress and apoptosis.The differentiation of human umbilical cord mesenchymal stem cells(h UCMSCs)for tissue regeneration and repair has become a hot topic in regenerative medicine.HUCMSCs are easy to obtain and collect,easy to culture,and have strong proliferation ability in vitro.Some studies have shown that umbilical cord mesenchymal stem cells are more capable of osteogenic differentiation than other types of stem cells.The clinical trials and applications of h UCMSCs in bone tissue engineering have great potential.To explore the inducers for inducing osteogenic differentiation of h UCMSCs will lay a foundation for its wide application.First,low-dose AS-IV 10mg/ m L(AS-IV-L group),medium-dose AS-IV 50mg/ ml(AS-IV-M group)and high-dose AS-IV 100mg/ ml(AS-IV-H group)were prepared.CCK-8 assay was used to detect the proliferation of h UCMSCs in vitro at different concentrations of astragaloside IV.With the increase of AS-IV concentration,the activity of h UCMSCs increased.h UCMSCs osteogenic differentiation was induced for 21-28 days,and alizarin red staining analysis was performed.Finally,Image J software is used to calculate the gray value.In molecular experiments h UCMSCs were divided into Control group,AS-IV-L group,AS-IV-M group and AS-IV-H group.After 48 h and 72 h of processing,the original data were analyzed by high-throughput sequencing.Western Blot was used to verify the promoting effects of different concentrations of AS-AV on RUNX2,OPN and PAPP-A2.RT-PCR was used to verify the effect of IGF-1/IGF-1R signaling pathway on h UCMSCs.In this paper,the effect of AS-IV on the osteogenic differentiation of umbilical cord mesenchymal stem cells was investigated by high-throughput sequencing.The molecular mechanism is clarified at the molecular level with the development of modern scientific means.It provides theoretical basis and experimental experience for the modernization of Chinese medicine to the world.
