Study On Cynanchum Otophyllum And Its Effective Compounds Group Combined With Phenobarbital On The Antiepileptic Effect

Effective compounds group of Chinese materia medica is the main material basis for efficacy in Chinese materia medica,which plays a core role in Chinese materia medica formula.Cynanchum otophyllum(QYS)is a national medicine in the southwest region,and now it has been developed into the tablets,has anti epilepsy with western medicine,the treatment of refractory epilepsy,the clinical effect is exact.At the same time,it can reduce the liver damage,drowsiness and other side effects by phenobarbital medicine.In the preliminary work of our laboratory,we screened out the compounds with anti epileptic activity,including M1 and Otophylloside N,which have anti epileptic activity through the digestion stability test and serum drug chemistry test.Purpose of study:This paper will summarize the successful case study of the existing natural medicine effective components group through literature review.QYS as an example,and combined with phenobarbital(PHB)anti epilepsy disease.The evaluation of bioequivalence evaluation was carried out from pharmacokinetic(PK)and pharmacodynamic study(PD)to explore the relationship between effective components(M1 and OtoN),the effective components group(M1+OtoN)and the overall system(QYS),environment(PHB).We tried to explore a new path for the study of the effective components of natural medicine.Main research contents:1,The successful case study and discussion.2,Content determination and separation.3,Establish the determination method.4,Construction of the effective components group.5,Pharmacodynamics.6,pharmacokinetics.Conclusion:1,Impact from phenobarbital on QYS:QYS can enhance the phenobarbital on antiepileptic and the onset time advanced and the duration lengthened.In addition,it protects the liver and reduces the concentration of phenobarbital in brain.It was accordance with the clinic that QYS can use with phenobarbital on antiepileptic and can reduced the toxin.The data of PK showed that this phenomenon was related to the AUC and Cmax of phenobarbital.2,Impact from QYS on phenobarbital:Phenobarbital can enhance the QYS and its effective(group)on antiepileptic and the onset time advanced and the duration lengthened.It was accordance with the clinic that QYS tablet can use with other drugs on antiepileptic.The data of PK showed that this phenomenon was related to the AUC and Cmax of M1 and OtoN.Phenobarbital can enhance QYS on antiepileptic but the AUC and Cmax of M1 and OtoN in it were reduced.This may related to the complex content of QYS.3,The relationship between system and the main factors:The speed of M1 and OtoN to into the body in Gro M1 and Gro OtoN was lower than Gro QYS and part lower than Gro M1+OtoN,Gro OtoN was more effective than Gro QYS and Gro M1+OtoN on antiepileptic.But M1 was great on protect the liver.4,The relationship between overall system and subsystem:The subsystem was more effective than the overall system on antiepileptic combined with phenobarbital.It has earlier onset time and more duration.And at the same time,the liver body ratio was lower.In addition,the content of Ml and OtoN in body was lower and consumed little.
